优裕环境胎教与地震恐惧胎损的SD大鼠母子两代行为遗传学研究

摘要
目的:研究母鼠优裕环境胎教,地震恐惧胎损以及补肾中药对子鼠生长发育、行为表征的影响。方法:将16只受孕SD母鼠按照随机分为4组:优裕"胎教"组(优裕组)、空白对照组(对照组)、地震模拟"胎损"组(地震组),地震模拟胎损后中药干预组(中药组)。观测存活子鼠的体重、身长、尾长、空格试验、睁眼情况、亮盒排便、翻身测试。结果:①优裕环境、地震模拟以及地震模拟后中药干预对子鼠的体重、身长、尾长的影响随时间变化的趋势皆不一样,且具有统计学意义(P<0.05);②在空格试验中中药组空格数大于其余三组,且差异具有统计学意义(P<0.05),但优裕组与对照组空格数无统计学差异,地震组与对照组亦无统计学差异;③四组睁眼率差异具有统计学意义(χ2=16.402,P=0.001<0.05),但亮盒排便测试与翻身测试各组间均无统计学意义。结论:优裕环境母鼠胎教可以增加子鼠的体重、身长从而促进子鼠的生长发育;母代的地震恐惧影响可以遗传到子代,并降低体重和身长的延缓子代的生长发育;金匮肾气丸可纠正地震恐惧导致的子鼠发育迟缓。
Objective:It is aimed to study the hereditary effects of fetus education from rich environment,fetus injury from earthquake simulation and kidneys-supplementing herbs of pregnant SD rats on their offspring.Methods:16 pregnant rats were randomized to four groups:rich environment group,control group,earthquake group and herb group.The body weight,body length,tail length,spaces test,eyes-opening,light box defecation test,and turn-over test of survived offspring were measured and observed.Results:(1)Compared to the control group,the alteration trends along with the growth of the offspring of the influences that the rich environment,earthquake simulation and herbal intervention after earthquake simulation brought to the body weight,body length and tail length of the offspring,were statistically different(P<0.05).(2)In spaces test,child rats of the herb group performed more spaces than another three groups(P<0.05),but there was no significant difference between the rich environment group and the control group,the earthquake group and control group.(3)The eyes-opening rates of the four groups were statistically different(χ2 =16.402,P=0.001<0.05);there were no statistical difference among the groups in light box defecation test and turn-over test.Conclusion:Rich environment fetus education could improve the body weight and body length of the offspring so as to promote the growth of the offspring.The fear of pregnant rats derived from earthquake simulation could pass down to their offspring and reduce the body weight and body length and accordingly retard the growth.Jingui Shenqi Pill could correct the retardation of the offspring due to the fear from earthquake simulation.
