We compared different methods for the separation of positional isomers of PEG-rhGH. We developed a novel and effective method of IEC-HPLC by pH gradient elution to separate positional isomers of PEG-rhGH. Some essential characterizations of positional isomers were described. We successfully identified the PEGylation sites of PEG-rhGH and analyzed its spatial structure.
