Circulating cell-free nucleic acids (cfNAs) are readily taken up by healthy cells. The uptaken cfNAs activate DDR and integrate into host cell genomes. Illegitimate genomic integration of cfNAs leads to dsDNA breaks and apoptosis. Consequently, cfNAs act as ubiquitous, physiological, endogenous mutagenic agents. Progressive cfNAs-induced mutagenesis may be implicated in ageing and cancer.
