摘要
目的:应用数据挖掘方法对国家专利数据库的治疗心律失常中药的配伍规律进行研究,为临床治疗与研发中药新药提供参考。方法:检索国家专利数据库治疗心律失常的方剂发明专利295条,应用中医传承辅助平台(V2.5),行证候分布统计、药频统计,并运用关联规则、改进的互信息法、复杂系统熵聚类、无监督的熵层次聚类进行数据分析。结果:频次前4位的证候为气郁络阻证(132,44.75%)、阴阳两虚证(110,37.29%)、心神不宁证(74,25.08%)、气阴两虚证(45,15.25%);频次前4位的高频单药为甘草(132,44.75%)、丹参(103,34.92%)、麦冬(99,33.56%)、酸枣仁(92,31.19%);高频对药前3位为麦冬-五味子(49,16.61%)、麦冬-丹参(45,15.25%)、丹参-当归(40,13.56%);高频角药前2位为麦冬-丹参-五味子(25,8.47%)、炙甘草-麦冬-桂枝(22,7.46%);关联度最高的配伍为牡蛎->龙骨(置信度0.840),与其他药物关联度最高的单药为丹参;基于熵方法提取核心组合10组,候选新方5条。结论:现代临床治疗心律失常当以滋阴通阳复脉为法,兼以理气通络,用药可优先考虑炙甘草、丹参、麦冬、酸枣仁等,配伍可优先考虑龙骨配伍牡蛎等,核心组合可优考虑泽泻、琥珀、益母草等,候选新方可优先考虑远志、柏子仁、酸枣仁、白芍、白术、木香等。
        This study was aimed to analyze traditional Chinese medicine(TCM) prescription composing laws in treating arrhythmia from the China patent database(CNPAT) based on data mining technology in order to provide references for clinical treatment and new TCM prescription development. TCM prescription invention patent in treating arrhythmia was searched in CNPAT. A total of 295 prescriptions of syndrome and drug frequency were received. The TCM Inheritance Support System(V2.5) was used. The statistics was made on syndrome distribution and drug frequency. Association rules,improved mutual information method, complex system entropy clustering analysis and unsupervised entropy hierarchy clustering were adopted. The results showed that the top four high-frequency syndrome are qi stagnation and stasis among collaterals(132, 44.75%), deficiency of both yin and yang(110, 37.29%), heart and spirit uneasy(74, 25.08%),and deficiency of both qi and yin(45, 15.25%). The top four high-frequency single-drugs are glycyrrhiza(132, 44.75%),salvia(103, 34.92%), liriope(99, 33.56%), spine date seed(92, 31.19%). The top three high-frequency pair-drug combinations are liriope-schisandra(49, 16.61%), liriope-salvia(45, 15.25%), salvia-angelica(40, 13.56%). The top two high-frequency triangle-drugs combinations are liriope-salvia-schisandra(25, 8.47%), honey prepared glycyrrhizaliriope-cassia twig(22, 7.46%). The highest frequency correlation is oyster-> dragon's bones(The confidence range was0.840.). Salvia has the highest degree of association with other drugs. The 10 core combinations and 5 candidate new prescriptions were obtained based on entropy method. It was concluded that the TCM treatment method of replenishing yin, activating yang and recovering pulse can be used to treat arrhythmia in modern TCM clinical practice, in combination with regulating qi and removing stasis in collaterals. Single drugs such as honey prepared glycyrrhiza, salvia,liriope, spine date seed and so on can be first considered to be used. Drug combinations such as oyster-dragon's bones and so on can be first considered to be used. Core combinations such as alisma orientale, amber, leonurus and so on can be first considered to be used. Candidate new prescriptions such as milkwort, seed of oriental arborvitae, spine date seed,white peony root, atractylodes, radix aucklandiae and so on can be first considered to be used.
