摘要
选用乙醇作为沉淀剂,对鞘氨醇单胞杆菌Sphigomonas sp.WG产威兰胶的提取工艺进行设计和优化。考察了发酵液加热处理、固液分离方式、乙醇用量、沉淀温度和沉淀时间对威兰胶提取量的影响,同时研究了3种不同干燥方式对威兰胶溶液粘度的影响。优化后的威兰胶醇沉工艺为:发酵液与乙醇的混合比例为1∶5.5(体积比),乙醇沉淀温度和时间分别为20℃和6 h,威兰胶沉淀物采用过滤分离,最后经真空冷冻干燥获得成品。
        The extraction process of Welan gum from Sphigomonas sp. WG was designed and optimized with ethanol as a precipitator. The influences of fermentation liquor heating treatment, solid-liquid separation method, ethanol dosage, precipitation temperature and time on Welan gum yields were investigated. In addition, the effects of three drying methods on the viscosity of Welan gum were also examined. The optimized extraction process was set as follows. The mixing ratio between fermentation liquor and ethanol was 1∶5.5(volume ratio), the precipitation temperature and time were 20 ℃ and 6 h respectively. Welan gum precipitation was separated by filtration, and the finished product of Welan gum was obtained by vacuum freeze-drying the samples.
