BMSCs移植联合电针刺激对ICH大鼠GAP-43表达的影响

摘要
目的探讨BMSCs移植联合电针刺激对脑出血(ICH)的作用及其对GAP-43表达的影响。方法将GFP标记的BMSCs常规复苏、培养和传代;利用脑立体定位仪在SD大鼠脑尾壳核内注射胶原酶和肝素构建ICH模型;ICH模型大鼠随机分为对照组(NS组)、BMSCs移植组(BMSCs组)和BMSCs移植联合电针组(Ea-BMSCs组),分别将生理盐水和BMSCs注入模型鼠尾壳核内,并用电针刺激Ea-BMSCs组大鼠百会穴和大椎穴,在不同时间点对大鼠行神经损害程度评分(NSS评分),采用RT-PCR和Western blot检测各组大鼠脑组织GAP-43m RNA及蛋白的表达情况。结果造模后移植前各组为脑中度损伤,NSS评分一致,无统计学差异(P>0.05);移植后第5天和7天BMSCs组与NS组比较NSS评分显著下降(P<0.05);第5天、7天和14天Ea-BMSCs组分别与BMSCs组和NS组比较NSS评分下降明显(P<0.05);移植后第5天、7天和14天,Ea-BMSCs组和BMSCs组GAP-43 m RNA及蛋白表达水平较NS组水平显著升高(P<0.05);而在第7天和14天Ea-BMSCs组较BMSCs组表达更强(P<0.05)。结论BMSCs移植联合电针刺激较单纯BMSCs移植能更有效促进ICH大鼠脑组织GAP-43表达,进而促进轴突生长,有利于神经组织重塑。
Objective To explore the effect of BMSCs transplantation combined with electroacupuncture stimulation on intracerebral hemorrhage(ICH) rats as well as the expression of growth associated protein-43(GAP-43).Methods After routinely recovered,GFP-labeled bone mesenchymal stem cells(BMSCs) were inductively cultured and passaged.ICH models were established by the injection of collagenase and heparin in SD rats' caudate nucleus through using brain stereotaxic instrument.ICH rats were randomly divided into control group(NS group),BMSCs transplantation group(BMSCs group) and BMSCs transplantation combined with electro-acupuncture group(Ea-BMSCs group),with respectively cerebral infusion NS and BMSCs as well as supplement with electro-acupuncture stimulation in rats Baihui and Dazhui in Ea-BMSCs group.The damage of rat nerve was assessed with NSS score system at different time points.Meanwhile,GAP-43 expression was detected by RT-PCR and western blot analysis.Resμlts After modeling and before transplantation,ICH rats in different groups displayed moderate cerebral injury and NSS scores were nearly identical without significant difference(P>0.05).Compared with NS group,NSS scores in BMSCs group apparently decreased in 5 day and 7 day after transplantation(P<0.05).However,NSS scores of Ea-BMSCs group were significantly lower than those of BMSCs group and NS group in 5 day,7 day and 14day(respectively P<0.05).The levels of GAP-43 m RNA and protein expression in Ea-BMSCs group and BMSCs group both remarkably enhanced when compared with those in NS group in 5 day,7 day and 14 day after transplantation(respectively P<0.05).Furthermore,GAP-43 m RNA and protein expressions in EaBMSCs group were higher than those in BMSCs group in 7 day and 14 day(respectively P<0.05).Conclusion BMSCs transplantation combined with electro-acupuncture stimulation can be more effective to promote GAP-43 expression in ICH rat than the single BMSCs transplantation,then enhance neurite growth and improve brain pathological structure.
