双氢睾酮诱导的多囊卵巢综合症合并动脉粥样硬化大鼠模型的建立

摘要
目的建立多囊卵巢综合症(Polycystic Ovary Syndrome,PCOS)合并动脉粥样硬化(AS)大鼠模型。方法通过高脂饲料喂养长期颈后皮下埋置双氢睾酮(DHT)大鼠的方式建立PCOS合并AS大鼠模型,证明此模型不仅具有与PCOS类似的代谢异常;还合并了血脂增高、动脉光镜病理形态改变等动脉粥样硬化性改变。结果 DHT埋置组食物摄取量、体重、生殖激素水平、卵巢滤泡、血脂、血糖及胰岛素水平的变化符合PCOS代谢方面变化;笔者给PCOS大鼠(DHT埋置组)模型给于高脂饲料,而且于造模后12周,管壁明显增厚,内膜明显增厚,16周内皮细胞损伤脱落,深层胶原纤维暴露,有单核细胞黏附,可见动脉粥样硬化斑块。结论通过高脂饲料喂养DHT诱导的PCOS大鼠,建立PCOS合并AS大鼠模型,再现了人类的典型临床症状及病理特征,是研究多囊卵巢综合症合并动脉粥样硬化可行的动物模型。
Objective To establish a rat model of polycystic ovary syndrome(Polycystic Ovary Syndrome,PCOS) with atherosclerosis(AS).Methods The clean grade SD rats were selected as the experimental animal,and the neck subcutaneous of rats were placed by dihydrotestosterone(DHT) with feeding by high fat diet for long-term,and the rat models not only has metabolic abnormalities similar with PCOS;but also the changes of blood lipids and arterial light microscopic pathology similar with atherosclerosis.Results Food intake,body weight,reproductive hormone levels,ovarian follicles levels,blood lipid levels,blood glucose levels and insulin levels in DHT-induced model in line with PCOS metabolic changes;Tube wall were markedly thickened in DHT-induced model by high fat feed after 12 weeks,and intimal wall were significantly thickened in 16 weeks,and endotheliocyte injury,deep collagen fiber,monocyte adhesion,visible atherosclerotic plaques were observed in this model.Conclusion DHT-induced PCOS rat model by high fat feed reproduces the human typical clinical symptoms and pathological characteristics,it can provide a relatively simple and feasible rat model for further study of the mechanism of PCOS with AS.
