电针对帕金森病小鼠模型脑nNOS和Calbindin-D28k表达的影响

摘要
目的观察电针治疗对帕金森病(PD)小鼠脑内神经元型一氧化氮合酶(n NOS)和钙结合蛋白D28K(Calbindin-D28k)的表达影响,以探讨电针防治PD的作用机制。方法采用1-甲基-4-苯基1,2,3,6四氢吡啶(MPTP)腹腔注射建立PD小鼠模型,电针"舞蹈震颤控制区"和"百会"穴,针刺疗程分别为7天、14天和28天,利用免疫组化技术观察对各组小鼠脑酪氨酸羟化酶(TH)阳性细胞、n NOS及Calbindin-D28k表达的影响。结果模型组小鼠黑质TH阳性神经元明显减少(P<0.001),而电针组14天、28天小鼠TH阳性神经元明显增多(P<0.05,P<0.001);模型组小鼠黑质和纹状体内n NOS的表达较正常组明显增多(P<0.01,P<0.001),电针组PD小鼠黑质n NOS的表达均明显减少(P<0.001),纹状体中只有电针28天小鼠n NOS的表达显著减少(P<0.001);模型组黑质Calbindin-D28k的表达明显减少(P<0.001),电针14天和28天组的表达较模型相比明显增多(P<0.01,P<0.05)。结论提示电针可能通过降低脑内n NOS的表达,增加Calbindin-D28k的表达,以提高PD小鼠脑抗氧化能力,达到保护黑质多巴胺能神经元、改善PD运动障碍的作用。
Objective To observe the effect of electroacupuncture on the expression of neuron nitric oxide synthase(n NOS)and Calbindin-D28 k in the brain of Parkinson's disease(PD) mice,and to explore the mechanism of electroacupuncture on PD.Methods Mice model was established by intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP),electroacupuncture at"dancing tremor control area"and"Baihui"was applied at"dancing tremor control area"and"Baihui",and immunohistochemical method was used to observe the effects on the tyrosine hydroxylase(TH) positive cells and the expression of neuronal nitric oxide synthase(n NOS) and calbindin-D28 k in each group.Resluts The number of TH-positive neurons in the Substantia nigra pars was significantly decreased in the model group at 7 days,14 days and 28days(p<0.001),while the TH-positive neurons in the electroacupuncture group increased significantly at 14 days and 28 days(p<0.05,p<0.001).The expression of n NOS in the substantia nigra and striatum of the three model groups was significantly higher than that in the normal group(P<0.01,P<0.001).In substantia nigra,electroacupuncture significantly decreased the expression of n NOS of PD mice in the three survival periods(P<0.001),but in the striatum,the expression of n NOS was decreased only at 28 days.The expression of Calbindin-D28 k in the model group was significantly lower than that in the normal group(P<0.001),and the expressions of the electroacupuncture groups at 7 days,14 days and 28 days was significantly higher than that of the model group(P<0.01,P <0.05).Conclusion By decreasing the expression of n NOS in the brain,the electroacupuncture may improve the expression of Calbindin-D28 k so as to increase the antioxidant capacity of PD mice,to protect the dopaminergic neurons in the substantia nigra and to alleviate the PD movement disorder.
