雷公藤甲素诱导HPV阳性肿瘤CaSki细胞凋亡的分子机制研究

摘要
目的:雷公藤甲素(Triptolide)是一种从中药植物中提取的环氧二萜内酯类化合物,一直以来被认为具有广谱的抗瘤活性,能够抑制多种肿瘤细胞的增殖并诱导其发生细胞凋亡。本文以HPV阳性的人宫颈癌上皮细胞CaSki为模型,验证了雷公藤甲素诱导其发生凋亡可能的机制。方法:本文采用MTT法,流式细胞技术以及PCR的方法对雷公藤甲素的抗肿瘤活性以及诱导肿瘤细胞发生凋亡进行了研究。结果:实验结果表明,雷公藤甲素对HPV阳性的人宫颈癌上皮CaSki细胞的生长具有抑制作用,IC_(50)为25.289±0.375nM。通过Annexin V-PI染色,流式细胞技术结果表明,雷公藤甲素能够诱导细胞发生凋亡。通过PCR发现雷公藤甲素能够抑制细胞p53的激活。结论:雷公藤甲素可能通过上调p53表达从而诱导HPV阳性人宫颈癌上皮CaSki细胞发生凋亡。
Objective:Triptolide is a diterpene lactone extracted from Chinese medicinal herb Tripterygium Wilfordii Hook F,which has been used to treat inflammatory and immunological diseases.Yet the anti-tumor activities of Triptolide has been found for several years.Therefore,the experiments have been designed to investigate the mechanism of the Triptolide to induce the apoptosis of HPV positive CaSki cells.Method:MTT method has been used to calculate the IC50 of Triptolide to Caski cells and the flow cytometry technique has been applied to determine the apoptosis occurredin Caski and to acquire the percentage of the apoptotic cells versus to normal cells.And western blot and PCR has been used to test the expression of protein and mRNA.Result:The IC50 of triptolide to Caski cells was 25.289 ±0.375 nM for 24 hours and the Annexin V-PI stain has shown that CaSki cells treated with Triptolide were induced to apoptosis and we found the P53 was up-regulated at transcription and protein level.Conculsion:We could conclude that P53 might be involved in the CaSki cells apoptosis induced by triptolide treatment.
