枳壳及其主要活性成分对脾虚模型大鼠血清胃泌素、血浆乙酰胆碱、胃动素、P物质和血管活性肠肽的影响

摘要
目的:研究枳壳水煎液及其主要活性成分对脾虚模型大鼠血清胃泌素(GAS)、血浆乙酰胆碱(ACh)、胃动素(MTL)、P物质(SP)和血管活性肠肽(VIP)的影响。方法:以苦寒泻下加饥饱失常法复制大鼠脾虚模型,枳壳水煎液及其主要活性成分分别灌胃给药后,采用ELISA法检测大鼠血清胃泌素、血浆乙酰胆碱、胃动素、P物质和血管活性肠肽的含量变化,观察各组给药后对脾虚模型大鼠胃肠激素的影响。结果:与空白对照组比较,模型组大鼠中ACh、GAS、MTL和SP含量显著降低(P<0.01),而VIP含量则显著升高(P<0.05)。各组给药后,柚皮苷低、中、高剂量组血浆中ACh含量,柚皮苷低、中剂量组血浆中SP含量和柚皮苷低剂量组中VIP含量与模型组比较显著降低(P<0.05,P<0.01),而柚皮苷中、高剂量组血浆中SP含量和柚皮苷低、中剂量组血清中GAS含量则较模型组显著升高(P<0.05,P<0.01);新橙皮苷低剂量组血浆中ACh含量较模型组显著降低,而新橙皮苷高剂量组血浆中ACh含量,新橙皮苷低、中、高剂量组血浆中SP含量,新橙皮苷中、高剂量组血浆中MTL含量和新橙皮苷低、中、高剂量组血清中GAS含量则较模型组显著升高;两个单体组合物低剂量组血浆中ACh含量和两个单体组合物低、中剂量组血浆中VIP含量与模型组比较显著降低,而两个单体组合物中、高剂量组血浆中ACh含量,两个单体组合物高剂量组血浆中SP含量,两个单体组合物高剂量组血浆中MTL含量和两个单体组合物低、中、高剂量组血清中GAS含量则较模型组显著升高;枳壳水煎液低剂量组血浆中ACh含量与模型组比较显著降低,而枳壳水煎液高剂量组血浆中MTL含量和枳壳水煎液低、中、高剂量组血清中GAS含量则较模型组显著升高。结论:枳壳可能通过升高血清中GAS、血浆中.ACh、MTL、SP含量和抑制VIP的分泌来促进胃肠运动,这可能是枳壳治疗胃肠动力障碍性疾病的机制之一。
Objective To study effect of Fructus Aurantii and its main active ingredients on gastrin in serum,acetylcholine,motilin,substance P and vasoactive intestinal peptide in plasma of spleen deficient model in rats.Methods:Spleen deficient model was induced by bitter-cold diarrhea and hunger full disorders replicate.By ELISA for detection of the content change serum gastrin,plasma acetylcholine,motilin,substance P and vasoactive intestinal peptide of rats,observed Fructus aurantii and its main active ingredients effect on rat model of spleen deficiency syndrome after orally administered.Results:The levels of ACh,GAS,MTL and SP increased(P<0.01),and VIP decreased significantly compared with normal group(P<0.05).After each group administration,the level of plasma for Ach in low,middle and high-dose group,plasma for SP in low,middle-dose group,and plasma for VIP in low-dose group of naringin decreased significantly compared with normal group(P<0.05,P<0.01).The level of plasma for SP in middle,high-dose group and serum for GAS in low,middle-dose group of naringin increased significantly compared with normal group(P<0.05,P<0.01).The level of plasma for Ach in low-dose group of neohesperidin decreased significantly compared with normal group.The level of plasma for ACh in high-dose group,plasma for SP in low,middle-dose group,plasma for MTL in middle,high-dose group and serum for GAS in low,middle-dose group of neohesperidin increased significantly compared with normal group.The level of plasma for Ach in low-dose group and plasma for VIP in low,middle-dose group of two monomer composition decreased significantly compared with normal group.The level of plasma for ACh in middle,high-dose group,plasma for SP in high-dose group,plasma for MTL in high-dose group and serum for GAS in low,middle,high-dose group of two monomer composition increased significantly compared with normal group.The level of plasma for Ach in low-dose group of Fructus aurantii water decoction decreased significantly compared with normal group.The level of plasma for MTL in high-dose group and serum for GAS in low,middle,high-dose group of Fructus aurantii water decoction increased significantly compared with normal group.Conclusion:Fructus aurantii mechanisms of adjusting the gastrointestinal may be closely related to increasing the levels of ACh,GAS,MTL and SP,and decreasing the levels of VIP,perhaps this is one of the mechanisms of Fructus aurantii treatment of gastrointestinal motility disorders.
