刀鲚生长抑素的分子克隆、组织表达及胚胎发育表达

摘要
生长抑素(somatostatin,简称SS)属于多基因家族,是一类在各组织中广泛存在的多功能肽。鱼类中含有多个生长抑素基因,可编码多种形式的生长抑素多肽与其受体特异性结合发挥作用,目前已在鱼类中分离克隆得到6种生长抑素基因和4种生长抑素受体基因。在本研究中,我们克隆了刀鱼SS1基因的全长,利用实时荧光定量PCR的方法研究了SS1基因的组织表达情况,结果表明SS1在脑中特异性表达,在心、肝和肾等代谢相关组织中表达量稍高,在其他组织中表达微弱。SS1基因在胚胎发育早期即被检测到,随着幼体发育其表达量先是有所降低,之后逐渐升高并保持稳定。本研究显示了SS1基因在生长发育、代谢调节中的潜在作用,为刀鱼SS1基因的进一步研究奠定了基础。
Somatostatin(SS) is a kind of multifunctional tetradecapeptide widely distributed in various tissues, it belongs to multigene family. There are different somatostatin genes can encode multiple forms of somatostatin peptide playing roles by binding with somatostatin receptors distinctly in fish. To date, six somatostatin genes and four somatostatin receptors have been characterized in fish. In this study, highly conserved SS1 cDNA was characterized in Coilia nasus.The tissue distribution of C. nasus SS1 mRNA was analyzed by real-time PCR, demonstrating high level of expression in the brain, low levels comparatively in the heart, liver and kidney but with weak distribution in other tissues. During embryogenesis, C. nasus SS1 mRNA was detected in early-stage developing embryos and was observed to decrease firstly then increase to a stable level throughout larval development. This study has revealed potential role for SS1 in regulation of growth, development and metabolism, and provides a basis for further study of SS1 in C. nasus.
