仿刺参补体类似物AjC3、AjC4化学发光免疫检测方法研究

摘要
本实验应用吖啶酯(acridinium ester,AE)标记抗体,采用双抗体夹心化学发光免疫定量分析方法对仿刺参体腔液补体类似物等免疫因子进行定量检测。建立了补体化学发光免疫检测标准曲线,补体C3线性方程为:y=867.89Ln(x)+8963,相关系数为0.9949;补体C4线性方程为:y=3716.6Ln(x)+67101,相关系数为0.9970;测定结果显示,建立的检测方法能够检测到仿刺参体内补体类似物,仿刺参体腔液中补体类似物AjC3平均含量为4500.485±1610.71ng/ml;AjC4平均含量为188.53±36.58ng/ml,检测限值可达10-14g/ml。
Chemiluminescent immunoassay(CLIA) method was developed for quantitative detection of complement analogues, AjC3, AjC4 in Apostichopus japonicus for the first time.Double-antibody labeled by acridinium ester of chemiluminescent immunoassay was developed The complement test standard curve of Chemiluminescent immunoassay was established. The C3 linear equation was: y = 867.89Ln( x) + 8963, correlation coefficient was 0.9949, and the C4 linear equation was: y = 3716.6Ln(x)+ 67101, correlation coefficient was 0. 0.9970, The results showed that the complement analogues, AjC3, AjC4 in Apostichopus japonicus can be detected by the method, and the average content of AjC3 was 4500.485±1610.71ng/ml, and AjC4 was188.53±36.58ng/ml, The detection limit was 10-14g/ml.
