蔗糖酯型阳离子类脂的设计合成及其体外基因转运研究

摘要
基因治疗已成功用于癌症、HIV等疾病的治疗,其中获得高效低毒基因载体是关键。阳离子脂质体作为基因载体中的一种,具有制备方法简便、重复性好、生物降解性好和易于商品化等优点,目前已经越来越受到科研人员的关注~([1])。但现有阳离子脂质体还存在转染效率较低和一定的细胞毒性~([2])。蔗糖酯良好的生物相容性和降解性为克服传统阳离子类脂较高的毒性提供了解决途径,由于单一结构蔗糖酯难以化学合成,目前尚未有蔗糖酯型阳离子化合物用于基因转运研究。针对上述问题,本文设计合成了以季铵盐为头部,蔗糖为骨架,酯为连接键,不同长度的烷基链(C_(12)、C_(14)和C_(16))为疏水尾部的蔗糖酯型阳离子类脂,将其构建成阳离子脂质体,作为基因载体用于体外转运研究,并取得较好的体外转染效果和较低的细胞毒性。
Cationic lipids have been favored for many potential advantages compared with other non-viral vectors, including significant simplicity and ease of production, good repeatability and biodegradability, potential commercial value, and wide range of clinical application and safety. However, the problem which cationic liposome was lower transfection efficiency and higher cytotoxicity remains formidable challenges. In this article, cationic lipids containing quaternary ammonium salt headgroup, the ester linker, the sucrose backbone and different hydrocarbon tails(C_(12), C_(14) and C_(16)) were designed and synthesized. These cationic liposomes were used for gene delivery and obtained higher transfection efficiency and lower cytotoxicity.
