高通量测序技术在饮用水微生物多样性研究中的应用

摘要
微生物在饮用水处理过程中的作用是非常重要的,尤其生物滤池中的微生物对饮用水有机物去除起到了关键作用。而对该环境中的微生物群落结构及多样性缺乏系统全面的研究,主要是采用传统的分离培养、DNA指纹图谱技术、磷脂脂肪酸分析法及微阵列基因芯片等技术,但存在通量低和信息量小等缺点。随着第二代高通量测序技术的发展,克服了以上缺点,对于我们全面、准确和快速地了解生物滤池中的微生物群落结构及功能多样性提供了一个很好的手段。为此,本文在重点介绍第二代高通量测序技术,包括测序仪原理、类型及优越性,并在此基础上综述了该技术在分析饮用水环境微生物群落多样性中的应用。同时得出了高通量测序技术在饮用水环境微生物群落的多样性研究中具有很大的应用价值,为生物处理技术在饮用水生产中的更好应用奠定了坚实的理论基础。
Indigenous bacterial communities are essential for drinking water treatment,especially in biofilters for organic mattersremoval,yet microbial community structure remains poorly characterized.Therearemanymethodshavebeenusedforanalyzing microbial diversity,including traditional isolation and culture methods,DNA fingerprintion,phospholipid fatty acid analysis and gene-chip.However,these methodssufferfromseveraldrawbackssuchas low through-put and less information.High throughput sequencingprovides agood tool to systemataciallyanalyzemicrobial community structure in biofilters.Therefore,the aim of this study is to introduce the principle and advantages of high throughput sequencing and sequencertype,and summarize the application of high throughput sequencingin analyzing microbial community diversity in drinking water environment.Meanwhile,weconclude that high throughput sequencingis of great value in application in studying microbial community diversity of drinking water,which lay a solid theoretical basis to make biological treatment technologybetter apply in drinking water treatment process.
