阳离子脂质体与基因复合物的结合动力学

摘要
基因治疗作为一种全新的医疗手段,其有效性已得到临床试验的验证。基因递送载体技术是基因治疗的核心技术之一。阳离子脂质体作为一种重要的非病毒基因载体,在基因治疗领域具有广泛的应用前景。带正电荷的阳离子脂质体通过静电吸引作用与带负电荷的DNA结合,形成脂质体/DNA复合物。阳离子脂质体有效地与DNA结合是实现基因高效转运的必要条件,但另一方面,脂质体与DNA结合过于紧密,又不利于在细胞内载体对DNA的释放。本实验利用琼脂糖凝胶电泳技术检测脂质体与DNA的结合能力,通过荧光共振能量转移技术,即以上转换纳米晶与脂质体结合作为能量供体,以染料标记的探针DNA作为能量受体,建立荧光共振能量转移体系,利用荧光光谱测量体系的发光强度,研究脂质体与DNA的结合动力学,揭示阳离子脂质体和DNA结合的紧密程度与基因转运效率之间的内在联系。
As an important non-viral gene vector,cationic liposome had shown a potential application in gene transfer and disease treatment.It was necessary to combine cationic liposome with DNA efficiently for gene transfer.However,it was not conducive for intracellular release of DNA if cationic liposome was combined with DNA too tightly.In this study,agarose gel electrophoresis was used to evaluate the DNA binding ability of liposome.Fluorescence resonance energy transfer technology and fluorescence emission intensity spectrum were used to inspect the binding kinetics of cationic liposome and gene complexes.To establishmented of fluorescence resonance energy transfer system,upconversion nanoparticles combining with cationic liposome were acted as energy donor,and the labeled DNA was energy receptor.The luminescence intensity of fluorescence spectrum measurement system was used to study the binding kinetics of cationic liposome and gene complex.
