野生百合花器官愈伤组织诱导及生根研究

摘要
以野生岷江百合、淡黄花百合、卷丹、宜昌百合、川百合、野百合以及东方百合杂种系‘西伯利亚'和‘索邦'的花丝和子房为外植体,以毒莠定(Picloram)为愈伤组织诱导剂,以吲哚丁酸(IBA)为生根剂,分别研究了不同浓度的Picloram和IBA对愈伤组织诱导和生根的影响。结果表明:花丝和子房为外植体都可诱导出黄色疏松的胚性愈伤组织,是愈伤组织诱导的良好材料;两个栽培品种比6个野生百合种更容易诱导出愈伤组织和生根;野生百合中,最高的平均诱导率发生在岷江百合花丝,培养基为MS+1.0mg/L Picloram,诱导率为79.3%±8.5%,子房的最高平均诱导率发生在野百合,培养基为MS+2.0mg/L Picloram,诱导率为78.0%±8.3%;最高平均生根率发生在野百合愈伤组织,培养基为MS+0.1mg/L IBA,生根率为79.5%±4.8%。
The filament and ovary of wild Lilium species,including L.regale,L.sulphureum,L.lancifolium,L.leucanthum,L.davidii and L.brownii,and Oriental Lily Hybrids‘Siberia' and‘Sorbonne' were used as explants.Picloram and IBA were used for induction and rooting of callus,respectively.The effect of different concentrations of Picloram and IBA on callus induction and rooting were studied.The results indicated that the filament and ovary were excellent materials to successfully induce yellow and loose embryogenic callus.Two cultivars were easier for callus induction and rooting than six wild Lilium species.Of the six wild Lilium species,the maximum average induction rate(79.3%±8.5%) occurred in L.regale filament and the used medium was MS+1.0mg/L Picloram.The maximum average induction rate(78.0%±8.3%)of ovary occurred in L.brownii and the used medium was MS+2.0mg/L Picloram.The highest average rooting rate(79.5%±4.8%) was found in callus of L.brownii and MS+0.1mg/L IBA was used as medium.
