耐冬山茶染色体核型分析

摘要
耐冬山茶[Camellia japonica(Naidong)]作为山茶属最北缘分布种群,具有极强的耐寒性,是园林中不可多得的花卉种质资源。本文以耐冬山茶根尖为试材,用染色体常规压片法对耐冬山茶进行了核型分析,并对其染色体制片技术的取样时间、解离时间进行了研究,结果表明:耐冬山茶根尖取样时间以上午11:00~12:00为佳,在1mol/L盐酸60℃恒温水浴中解离12min,改良石炭酸品红染色液染色10min,制片效果最好。耐冬山茶染色体数目为2n=2x=30,其核型公式为2n=2x=30=20m(2sat)+6sm(2sat)+4st,最长与最短染色体长度比为2.25,平均臂比值为1.72,臂比值大于2:1的染色体占全部染色体的比例为20%,属于2B型,核型不对称系数(As.K.%)为60.73%。
Camellia japonica(Naidong) as the northern margin of the distribution of Camellia,has extremely cold-resistance.It is a rare flower germplasm resources in the garden.The roots of Camellia japonica(Naidong) were used as experimental materials to study the karotypes analysis with conventional pressed disc method.And the sampling time and hydrolysing time on the chromosome squashing techique were also studied.The main results were as follows:The optimal sampling time of roots was11:00- 12:00am,and the roottips were hydrolysed with lmol/L HCL for 12 min at 60 t and stained with modified phenol fuchsin solution,With this method a very good effect could be obtained.The chromosome number of Camellia japonica(Naidong) was 2n = 2x = 30,and the karotype formulae was 2n = 2x = 30 = 20m(2sat) + 6sm(2sat) + 4st.The ratio of the longest chromosome to the shortest one was 2.25,the mean ratio of arm was 1.72,and the arm ratio of chromosome more than 2-1accounted for approximately 20%of all arm ratio of chromosomes.The type of the karotype belonged to 2B.The asymmetrical karyotype coefficient was(As.K.%) was 60.73%.
