固相支撑-液液萃取技术在生物分析样品前处理中的应用

摘要
固相支撑-液液萃取技术(SLE)充分发挥硅藻土多孔性、大比表面、低表面活性等特点,实现微观液液萃取,与传统LLE应用领域相似,适合由生物体液内萃取非极性和中等极性目标物,可极大去除内源性干扰物质,并可实现高通量样品操作,是临床生物分析实验室日常样品前处理的一种高效样品前处理工具。本文采用Cleanert SLE 96孔萃取板对血浆中替米沙坦进行萃取,LC-MS/MS定量分析,并考察SLE去除磷脂功能,结果表明SLE试验重现性良好,RSD小于4.36%;经LLE处理的样品,磷脂峰面积为SLE法70倍之多;LLE试验所需试验操作时间为SLE的6倍。对于生物样品的高通量LCMS/MS分析,SLE是一个能够取代传统LLE的选择。
Supported liquid extraction(SLE) could purify biological fluid sample effectively,the sorbent is highly purified diatomaceous earth with property of porosity,high specific surface area and low surface activity.SLE could replace most of LLE application by the similar principle,and suitable for extracting nonpolar and medium polarity compounds from biological fluids.SLE could eliminate most of endogenous phospholipid and protein which cause matrix effect,and could be used in daily sample pretreatment with the convenience of automation and highly throughput.This application employs Cleanert SLE plate to extract telmisartan from plasma and determine by LC-MS/MS.The results show excellent reproducibility of SLE with RSD less than 4.36%.In the comparison experiment of SLE and LLE,operate time of SLE is shorten 6 times to LLE,with 70 fold decrease of phospholipid area.SLE is an ideal choice for bio-analysis sample preparation.
