食源性功能多肽的定量构效关系研究与数据库构建

摘要
蛋白质水解产物中含有丰富的功能多肽,其具有抗氧化、抗菌、抗高血压、免疫调节、抗血栓等功能。我们广泛收集了文献以及数据库中报道的各种食源性的功能多肽序列与活性数据,采用2D与3D结构表征技术,结合分子模拟研究了各种多肽与其相应受体的作用动力学特征,分别应用线性与非线性的建模方法,包括偏最小二乘与支持向量机等,建立了各种功能多肽的定量-序列-结构-动力学-活性特征关系模型。进而分析了各种功能多肽的氨基酸序列以及其结构与生理活性之间的关系。在此基础上,我们构建了开放的食源性功能多肽数据库。研究结果益于深入理解各种食源性功能多肽的作用机制,亦可为生理活性肽在保健食品领域内的开发与应用提供相应的理论基础。
Protein hydrolysates contain abundant functional polypeptides which have multifaceted properties,such as antioxidation,antimicrobial,antihypertensive,immunomdulant and antithrombotic,etc.We collecthundreds of sequences and activities of functional polypeptidesfrom literature as well as database.Sequences and structures of these peptides are characterized by 2-D and 3-D structural methods,combing with the dynamic characteristics of various types of polypeptides withtheir corresponding receptorsby molecular simulation.We then utilize linear(partial least squares) and nonlinear(support vector machine) modeling methods to establish models of quantitative sequence-structure-dynamics-activity relationship of functional polypeptides,thereby to explore their sequence-structure-activityrelationship.On the basis above,we constructan opened database of foodderived functional polypeptides.Our results are beneficial to understanding of mechanism of food-derived polypeptides,also can provide the corresponding theoretical basis for development and applications offunctional peptides in the field of health foods.
