苦瓜粉对肥胖大鼠肝脏脂代谢的干预作用研究

摘要
目的:研究苦瓜粉对肥胖大鼠肝脏脂代谢的干预作用,并从苦瓜粉调节肝脏内质网应激的角度探索其作用机制。方法:通过连续8周饲喂SD大鼠高脂饲料的方法建立肥胖模型,分别灌胃苦瓜粉(400mg/kg bw)和水(4mL/kg bw)8周后,测定其肝脏脂质水平、肝脏抗氧化能力等指标,同时采用Realtime-PCR技术检测肝脏脂代谢关键基因和内质网应激标志物的变化。结果:与肥胖模型组相比,苦瓜粉可减缓大鼠体重增长速度,降低Lee's指数,增加其糖耐量;降低肝脏中总胆固醇(TC)、低密度脂蛋白(LDL)和丙二醛(MDA)的含量,增加高密度脂蛋白(HDL)含量,升高过氧化氢酶(CAT)、总超氧化物歧化酶(T-SOD)的活力;同时还能显著上调肝脏脂代谢基因(PPARα和CPT1α)的表达水平,下调内质网应激标志物(GRP78和CHOP)的表达。结论:苦瓜粉可有效改善肝脏的脂代谢异常,其作用机制可能与增强肝脏抗氧化能力、抑制肝脏内质网应激有关。
Objective:Study the effect of bitter melon powder(BMP) on liver lipid metabolism in high-fat diet(HFD)-induced obese rats.Methods:We induced the obese rat model by feeding high fat diet for 8 weeks,then treated these rats with BMP(400mg/kg bw) and water(4 ml/kg bw) respectively for 8 weeks,and finally determinated the levels of liver lipid,liver antioxidant capacity and others.The key genes of liver lipid metabolism and endoplasmic reticulum stress were assessed with Realtime-PCR.Results:BMP showed a higher anti-obesity activity with lower rate of increase of obese rats' body weights,decreasing the Lee's index,reversing HFD- induced glucose intolerance.BMP also decreased the hepatic levels of TC,LDL and MDA,increased the levels of HDL and the enzyme activities of CAT and T-SOD in liver.Moreover,BMP markedly enhanced the mRNA expression of PPARa and CPT1α,and decreased the mRNA expression of GRP78 and CHOP in liver.Conclusion:BMP can ameliorate the disorders of liver lipid metabolism,the mechanism may be related with BMP can strengthen the antioxidant abilityand inhibit the endoplasmic reticulum stress in liver.
