苦瓜改变肥胖大鼠肠道通透性与缓解炎症水平的相关性研究

摘要
目的:研究苦瓜粉(BMP)对肥胖大鼠炎症水平的缓解作用,并建立其与肠道通透性变化的关联。方法:以高脂饲料喂养SD大鼠建立营养肥胖模型,分别灌胃水[4 rmL/(kg bw)]和BMP[400mg/(kg bw)]8周,测定大鼠的体重、血糖、血清炎症因子(如:TNF-α、IL-6)和内毒素结合蛋白(LBP)等指标。通过Realtime-PCR(RT-PCR)技术检测近端结肠紧密连接蛋白ZO-1和Occludin mRNA的表达水平,并采用Aglient7890气相色谱测定大鼠尿液乳果糖/甘露醇(L/M)的排出比值。结果显示:BMP可明显减缓肥胖大鼠体重增速,降低大鼠的血糖、血清炎症因子和LBP的水平(p<0.05);显著上调近端结肠紧密连接蛋白ZO-1和Occludin mRNA的水平,分别是肥胖模型对照组的1.32倍和1.33倍(p<0.05);有效降低肥胖大鼠尿液中L/M比值(1.46±0.43 VS.0.90±0.41)(p<0.05)。结论:BMP可以改变肥胖大鼠的肠道通透性,其与BMP缓解炎症水平具有一定相关性。
Objective:Study the effects of bitter melon power(BMP) on the prevention of obesity-related inflammation and its potentialrelevance with gut permeability.Methods:Obese rats model was established by feeding high fat diet(HFD) and then gavaged with water[4 mL/(kg bw)]and BMP[400mg/(kg bw)]respectivelyfor 8 weeks.Finally,we determinated the levels of blood glucose,inflammatory cytokines,lipopolysac-charide-binding protein(LBP) by Elisa.Gut permeability was assessed by measuring mRNAexpression of tight-junction proteins(ZO-1 and Occludin) with RT-PCR and lactulose/mannitol(L/M) excretion ratio with Aglient 7890 gaschromatography.Results;BMP showed a higher anti-obesity activity with lower rate of increase of obese rats' body weights,significantlyreversingglucose intolerance,the levels of inflammatory cytokines andLBP.Morever,BMP markedly enhanced the mRNA expression of ZO-1 and Occludin(1.32 and 1.33 times than obese control,p<0.05),and decreased the ration of L/M(1.46 ±0.43 VS.0.90±0.41,p<0.05).Conclusion;BMP has benificial effects on the obesity-related inflammation,which is associated with the changes of gut permeability.
