摘要
目的:研究肿瘤相关抗原GCF2选择性剪接区在人肝癌细胞株、肝癌组织、癌旁组织及正常成人肝细胞及组织中的表达及意义。方法:采用RT-PCR检测GCF2 5个选择性剪接区(D1~D5)在人肝癌细胞株、43例肝癌组织、相对应的癌旁组织,以及正常成人肝细胞株及9例肝组织中的表达。结果:GCF2 4个选择性剪接区(D1~D4)在所检测的细胞株、肝癌组织、癌旁组织和正常成人肝组织中均未检测到表达;GCF2选择性剪接区D5仅在人肝癌细胞株检测到阳性表达;在43例肝癌组织及相应的癌旁组织中,有41例肝癌组织GCF2选择性剪接区D5呈阳性表达,阳性表达率为95.35%(41/43),而9例正常肝组织均未检测到GCF2选择性剪接区D5表达。结论:GCF2D5选择性剪接区在肝癌组织中有较高阳性表达,GCF2选择性剪接区D5可能与肝癌的发生发展有关。
        Objective:To study the GCF2 mRNA expression in 5selective shear zones of hepatocellular carcinoma(HCC)cell lines,liver tissues,the tissues adjacent to carcinoma,normal adult liver cells and tissues.Methods:RT-PCR was performed to detect GCF2 mRNA expression in 5choose shear zones(D1~D5)of GCF2 in HCC cell lines BEL-7404,HepG2 and SMMC-7721,43 HCC tissues and corresponding adjacent tissues,as well as the normal adult liver cell line HL-7702 and 9normal liver tissues.Results:Four selective shear zones(D1~D4)of GCF2 were not found in all the cell lines,liver tissues,the tissues adjacent to carcinoma and normal adults liver tissues above;D5selective shear zone of GCF2 only was found to express in all HCC cell lines above and most of HCC tissues,there were 41 HCC tissues to be positive expression of D5 selective shear zone of GCF2,the positive expression rate was 95.35%(41/43),and D5 selective shear zone of GCF2 was not found to express in 9normal liver tissues.Conclusion:The positive expression of D5 selective shear zone of GCF2 in HCC tissues and cell lines suggests D5 selective shear zone of GCF2 possibly be correlated to the development.
