活血散结中药复方含药血浆对PDGF干预下兔RPE细胞增殖及IGF-Ⅰ表达的影响

摘要
目的:探究血小板衍生因子(PDGF)促视网膜色素上皮细胞(RPE)细胞增殖的最佳浓度,并由此研究活血散结中药复方含药血浆对PDGF干预下兔RPE细胞增殖及胰岛素样生长因子-Ⅰ(IGF-Ⅰ)表达的影响。方法:成年健康家兔为对象,制备含药血浆。观察不同浓度PDGF对兔RPE细胞增殖的影响,选择最佳作用于兔RPE细胞的PDGF浓度。将实验分成空白对照组、正常血浆组、PDGF组、PDGF+AG1296组、PDGF+含药血浆组,观察活血散结中药复方含药血浆对PDGF干预下兔RPE细胞增殖的影响以及PDGF干预下兔RPE细胞IGF-ⅠmRNA和蛋白表达的影响。结果:选择10μg/L PDGF作为干预RPE细胞的最佳浓度。与PDGF组比较,PDGF+AG1296组、PDGF+含药血浆组细胞增殖显著抑制(P<0.01)。PDGF+AG1296组、PDGF+含药血浆组与PDGF组比较,IGF-ⅠmRNA和蛋白表达显著下调(P<0.01)。结论:活血散结中药复方含药血浆能抑制PDGF作用下兔RPE细胞的增殖。活血散结中药复方含药血浆作用于PDGF干预下所致PVR的细胞模型可抑制RPE细胞IGF-Ⅰ的表达。
        Objective: To explore the best concentration of PDGF to promote the proliferation of RPE cells. To study the effect of drug-containing plasma of Huoxue Sanjie Formula on rabbit RPE cell proliferation and IGF-Ⅰ expression under the PDGF intervention. Methods: Adult healthy rabbits were used to prepare drug-containing plasma. The effects of different concentrations of PDGF on the proliferation of rabbit RPE cells were observed. The PDGF concentration optimally acting on rabbit RPE cells was selected. The study would be divided into control group and normal plasma group, the plasma PDGF groups, PDGF+AG1296 group and PDGF+drug-containing plasma group. To observe effect drug-containing plasma of Huoxue Sanjie Formula on rabbit RPE cell proliferation and IGF-Ⅰ mRNA and protein expression. Results: 10μg/L PDGF was selected as the optimal concentration for intervening RPE cells. It had significant inhibition of cell proliferation in PDGF+AG1296 group and PDGF+drug-containing plasma group(P<0.01). Compared with PDGF groups, IGF-Ⅰ mRNA, IGF-Ⅰ protein expression in PDGF+AG1296 and PDGF+drug-containing plasma group significantly lowered, the difference had statistical significance(P<0.01). Conclusion:Drug-containing plasma of Huoxue Sanjie Formula can inhibit PDGF under the action of rabbit RPE cells proliferation and the expression of IGF-Ⅰ in RPE cells.
