美洲大蠊提取液对创面愈合中TGF-β1通路中Smad6、Smad9表达的影响及超微病理结构观察

摘要
目的:探索美洲大蠊提取液作用大鼠难愈合创面的生长优势及可能调控机制。方法:将造模成功后的大鼠按随机数字表法分为空白组、模型组、实验组、对照组,应用免疫组化观察Smad6、Smad9的表达;应用HE染色观察细胞病理形态;应用透射电镜观察细胞超微结构。结果:第20天时,与模型组、空白组、对照组比较,实验组Smad6表达显著下降,Smad9表达显著上升(P<0.05)。病理形态显示,实验组细胞炎性渗出、水肿明显,炎性反应进程迅速;透射电镜显示,实验组较模型组、对照组细胞结构连接紧密,线粒体数量增加(P<0.05)。结论:在TGF-β1通路中,美洲大蠊提取液可能通过下调Smad6,上调Smad9表达,加速创面细胞病理炎性反应进程,促进细胞线粒体、内质网、桥粒等的增生与分化,促进创面愈合。
        Objective: To explore the growth advantages and possible mechanism of on American cockroach extracting rat wounds with refractory healing. Methods: After successful modeling, rats were divided by random number table method into control group, model group, positive group and experimental group according to the balance. The expression of Smad6 and Smad9 was observed by immunohistochemistry. Cytopathological morphology was observed by HE staining. The ultrastructure of cells was observed by transmission electron microscopy. Results: The expression of Smad9 increased and the expression of Smad6 decreased in the experimental group, which was statistically significant compared with control group, model group,positive group on the 20 th day(P<0.05). Pathologically, the experimental group had obvious inflammatory exudation and edema in wound tissue, and the inflammatory reaction process was rapid, which was statistically significant compared with the other three groups. Compared with the model group and the control group, the experimental group had more closely connected and numerous cells, which had statistical significance(P<0.05). Conclusion: American cockroach extracting may effectively down-regulate the expression of Smad6 and up-regulate the expression of Smad9, accelerate the pathologic inflammatory response process of wound cells, promote the proliferation and differentiation of cell mitochondria, endoplasmic reticulum and desmosomes, and promote wound healing.
