兔包皮来源的上皮细胞与人来源脱细胞羊膜基质(HAAM)组织工程共同培养细胞层片的体外研究

摘要
目的探讨兔包皮来源的上皮细胞与人来源脱细胞羊膜基质(human acellular amniotic membrane matrix,HAAM)组织工程共同培养细胞层片的效果。方法将兔包皮来源的上皮细胞接种于实验组(脱细胞羊膜基质)和对照组(未脱细胞的羊膜上基质)上,待细胞在两种羊膜基质上生长至铺满表面,用活细胞示踪剂标记细胞,然后通过组织学观察上皮细胞在两种羊膜基质上的黏附及生长情况。结果包皮来源的上皮细胞在两组羊膜基质上均能增殖生长,细胞呈单层生长,活细胞示踪剂显示细胞生长良好,并具有活细胞功能。结论包皮来源的上皮细胞是良好的实验室组织工程化的种子细胞,脱细胞羊膜基质是良好的组织工程化尿道支架材料,该方法可成功培养上皮细胞层片,有望用于尿道重建。
        Objective To discuss the effect of composite culture of tissue engineered cell sheets with rabbit's foreskin-derived epithelial cells and human acellular amniotic membrane matrix(HAAM) in vitro. Methods Rabbit's foreskin-derived epithelial cells wereinoculated on human acellular/general amniotic membrane matrix and the cells were tracked with viable cell tracer when growing on the surface of the amniotic membrane matrix.Then the adhesion and growth of epithelial cells on two kinds of amnioticmatrix were observed histologically. Results Foreskin-derived epithelial cells grew well on HAAM with normal mono-layer morphology,which was proven by viable cell tracer. ConclusionsForeskin-derived epithelial cell is a desirable seed cell for urinary tract tissue engineering and grows well on HAAM.This method can successfully cultivate epithelial cell lamellae and is expected to be used for urethral reconstruction.
