摘要
目的探讨低蛋白条件下燃煤型氟中毒对雌鼠生殖功能的影响。方法选取120只断乳2周SD雌鼠,分对照组、低蛋白组、染氟组、低蛋白染氟组,每组30只;光镜下观察卵巢组织形态、各级卵泡数量变化;于不同时间段(60 d、120 d、180 d)动情期处死雌鼠,采集雌鼠血液分离血清,检测血清中雌二醇(E2)含量。结果成功建立动物模型;随着时间增加,低蛋白染氟组卵泡颗粒细胞轮廓较对照组略显模糊,且水肿现象较为严重,此外黄体和卵泡退化现象明显,成熟卵泡数量较低蛋白组和对照组高,60 d、120 d、180 d均有统计学差异(P<0.05),更高于染氟组,120 d、180 d有统计学差异(P<0.05),氟和蛋白对60 d、180 d闭锁卵泡的影响存在交互效应(F值分别为4.193,5.172,P均<0.05);低蛋白染氟组E2含量逐渐降低,60 d与180 d比较有统计学意义(P<0.05);氟和蛋白对60 d、120 d、180 d雌鼠E2的影响不存在交互效应(F值分别为0.119、2.552、1.836,P均>0.05)。结论低蛋白可加重燃煤型氟中毒对雌鼠卵巢生殖功能的毒性作用。
        Objective To investigate effect of fluorosis caused by coal burning on reproductive function in female rats under low protein conditions. Methods 120 female SD rats in 2 weeks of which weaning were randomly divided into 4 groups: control, low protein, high fluorine, low protein and high fluoride, 30 rats contained in each group. To observe the change of mor-phology and number of ovarian tissue follicles and corpus luteum under light microscope by HE staining. At different time(60 d, 120 d, 180 d) of the estrous female rats were executed, the blood was collected, separation of serum, the content of estradiol(E2) were measured in serum. Results Animal model established successfully; With the dose increased, low protein and high fluoride groups,the edema degree of granulosa cells was aggravated,cell structure and cell contours become blurred,atretic follicle and corpus luteum degradation gradually increased, the mature follicles occasionally. It gradually increased and higher than that of the control group and low protein group, 60 d, 120 d and 180 d were statistically significant(P<0.05),and higher than the high fluoride group,120 d and 180 d were statistically significant(P<0.05). Interaction of fluoride and protein have effects on 60 d, 180 d follicular atresia(F values were 4.193, 5.172, P<0.05), low protein and high fluoride groups of female rats the content of E2 decreased, 60 d and 180 d were statistically significant(P<0.05), the interaction of fluoride had no effect on 60 d, 120 d, 180 d of female rats E2(F value of 0.119, 2.552, 1.836, P<0.05,respectively). Conclusion Low protein can increase the coal burning fluorosis on reproductive function of female rats ' ovarian toxicity.
