紫癜性肾炎气虚血瘀证患儿舌苔菌群特异性菌属分析

摘要
目的:通过对紫癜性肾炎气虚血瘀证患儿和正常健康儿童舌苔菌群的相关性分析,寻找两者之间的差异菌属。方法:选取辽宁中医药大学附属医院2018年7月至2018年9月儿科住院确诊为紫癜性肾炎气虚血瘀证的患儿为病例组(14例,HSPN组,均为白厚苔),正常健康儿童为对照组(11例,HC组,均为薄白苔),采用16S rDNA高通量测序技术对舌苔菌群进行研究。结果:HSPN组患儿的OTU个数、Shannon指数、Ace指数和Chao指数显著低于HC组(P<0.05,P<0.01);HSPN组舌苔普雷沃氏菌属-7和奈瑟球菌属的丰度明显高于HC组;LEfSe多级物种差异分析发现共有25种差异菌;T-test统计分析结果表明HSPN组和HC组的毛绒厌氧杆菌属、弯曲杆菌属、Solobacterium、真杆菌属、念珠菌属、乳杆菌属、毛螺菌属和一种未确认的微生物种属含量存在明显的差异(P<0.05)。结论:与健康儿童的薄白苔相比,HSPN气虚血瘀证患儿白厚苔具有较低的菌群物种多样性,普雷沃氏菌属-7和奈瑟球菌属可能是HSPN气虚血瘀证患儿病理舌苔形成的关键菌属。
        Objective: To find out the specific bacterial genus in children with henoch-sch?nlein purpura nephritis(HSPN)with syndrome of qi deficiency and blood stasis. Methods: All the cases were obtained from hospitalized children with HSPN with syndrome of qi deficiency and blood stasis in the Affiliated Hospital of Liaoning University of Chinese Medicine between July2018 and September 2018. The children were divided into HSPN group(14 cases with thick white tongue coating) and healthy control group(HC group, 11 cases with thin white tongue coating). 16 S rDNA high sequencing technology was used to analyze the bacterial flora of tongue coating. Results: The amount of OTU, Shannon index, ACE index and Chao index were significantly lower in HSPN group than those in HC group(P<0.05, P<0.01). The relative abundances of Prevotella-7 and Neisseria in HSPN group were distinctly higher than those in HC group. There were 25 kinds of different levels of bacteria by LEfSe analysis.T-test analysis showed significant differences between two groups in the amounts of Lachnoanaerobaculum, Campylobacter,Solobacterium, Eubacterium, Candida, Lactobacillus, Lachnospira and one unidentified(P<0.05). Conclusion: Compared with thin white tongue coating of healthy children, the thick white tongue coating of HSPN children with qi deficiency and blood stasis syndrome have a lower diversity of tongue coating flora. Prevotella-7 and Neisseria may be the key bacteria for the formation of pathological tongue coating in HSPN children with syndrome of blood stasis due to qi deficiency.
