神经鞘磷脂酶在乳腺癌患者中的表达及与细胞免疫水平的相关性研究

摘要
目的探讨神经鞘磷脂酶在乳腺癌患者中的表达及免疫细胞调节作用机制。方法 2016年6月-2018年3月乳腺癌患者67例设为观察组;选择同期治疗的乳腺良性疾病患者56例设为对照组。采用免疫印迹法(Western blot)法完成观察组与对照组神经鞘磷脂合成酶SMS1和SMS2蛋白表达水平;采用流式细胞仪测定两组CD3+、CD4+、CD8+、CD4+/CD8+水平。结果乳腺癌组织中SMS1和SMS2阳性率与肿瘤大小、TNM分期及淋巴结转移差异有统计学意义(P <0.05);Western blot法结果表明:观察组乳腺癌组织中SMS1和SMS2蛋白相对表达量,高于癌旁组织与对照组(P <0.05);观察组CD3+、CD4+、CD4+/CD8+水平,均低于对照组(P <0.05);观察组CD8+水平,高于对照组(P <0.05)。结论神经鞘磷脂合成酶SMS1和SMS2水平在乳腺癌患者中呈高表达,与免疫细胞水平具有一定的相关性。
        Objective To investigate the expression of sphingomyelinase in breast cancer patients and the mechanism of immune cell regulation. Methods From June 2016 to March 2018, 67 patients with breast cancer were enrolled in the observation group. 56 patients with benign breast disease who were treated at the same time were selected as the control group. The expression levels of sphingomyelin synthase(SMS1 and SMS2) in the two groups were determined by Western blot. The levels of CD3+, CD4+, CD8+ and CD4+/CD8+ were determined by flow cytometry.Results The positive rates of SMS1 and SMS2 in breast cancer tissues were statistically significant(P < 0.05). The results of Western blot showed that the relative expression of SMS1 and SMS2 protein in breast cancer tissues was observed. It was both higher than the adjacent tissues and the control group(P <0.05). The levels of CD3+, CD4+ and CD4+/CD8+ in the observation group were lower than those in the control group(P <0.05), while the level of CD8+ in the observation group was higher(P <0.05). Conclusion The levels of sphingomyelin synthase, SMS1 and SMS2, are highly expressed in breast cancer patients and have a certain correlation with immune cell levels.
