摘要
目的建立HPLC法同时测定蒲公英中咖啡酸、绿原酸、菊苣酸和单咖啡酰酒石酸的含量,再以响应面法优化蒲公英的提取工艺。方法采用色谱柱Agilent Eclipse Plus-C18柱(250 mm×4.6 mm,5μm),流动相:乙腈(A)-0.4%磷酸水溶液(B)梯度洗脱,柱温:30℃,流速:1.0 mL·min~(-1),检测波长:323 nm,进样量:10μL。以料液比、提取时间和提取次数为自变量,以咖啡酸、绿原酸、菊苣酸和单咖啡酰酒石酸的提取率的总评归一值(OD)为因变量,结合响应面法优选最佳的提取工艺。结果确定提取工艺条件为加水倍数16倍,提取时间1 h,提取次数3次。结论本HPLC法适用于蒲公英中4种酚酸类成分的同时测定,响应面法可以对蒲公英提取工艺进行优化,方法简便,预测性良好。
Objective To establish a method for the simultaneous determination of caffeic acid, chlorogenic acid, cichoric acid and monocaffeyltartaric acid in dandelion by HPLC, and optimize the extraction process of dandelion by response surface methodology(RSM). Methods The samples were analyzed on Agilent Eclipse Plus-C18 column(250 mm×4.6 mm, 5 μm). The mobile phase was acetonitrile(A)-0.4% phosphoric acid aqueous solution(B) gradient elution, and the column temperature was 30 ℃. The flow rate was 1.0 mL·min~(-1).The detection wavelength was 323 nm, and the injection volumn was 10 μL. The optimum extraction process was optimized by RSM, with the ratio of material to liquid, extraction time and extraction times as independent variables and the total evaluation normalization(OD) of caffeic acid, chlorogenic acid, cichoric acid and monocaffeyltartaric acid as the dependent variables. Results The extraction condition was 16 times of water,extract for 1 h, and 3 extraction times. Conclusion The method is suitable for the simultaneous determination of 4 phenolic acids in dandelion. RSM can optimize the extraction of dandelion. The method is simple and predictive.
