摘要
目的探究二甲双胍是否可增强肝癌细胞对索拉菲尼的敏感性及其相关机制。方法 CCK-8及流式细胞术检测二甲双胍联合索拉菲尼处理或单一索拉菲尼处理后肝癌HepG2细胞的增殖活性和凋亡;Western blot技术检测凋亡相关蛋白及PI3K/AKT/mTOR通路的激活情况。结果较索拉菲尼组相比,二甲双胍+索拉菲尼组细胞增殖活性降低,凋亡上升,Bcl-2表达降低,而Bax、Caspase3表达升高,且p-PI3K、p-AKT及p-mTOR的蛋白表达水平升高。结论二甲双胍可增强肝癌细胞对索拉菲尼的药物敏感性,这可能与PI3K/AKT/mTOR通路和上皮间充质转化过程抑制相关。
        Objective To explore the effect and mechanism of metformin on sorafenib sensitivity of hepatocellular carcinoma cells.Methods CCK-8 and flow cytometry assay were used to assess cell proliferation and apoptosis after hepatocellular carcinoma HepG2 cells were treated with metformin+sorafenib or sorafenib;Western blot was used to test the expression of proteins related to cell apoptosis or to evaluate the activitaion of PI3 K/AKT/mTOR signaling.Results Compared with sorafenib,cell proliferation was inhibited,cell apoptosis was enhanced,the expression of Bcl-2 was decreased while Bax and caspase3 expression was elevated in metformin+sorafenib group,as well as increased expression of p-PI3 K、p-AKT and p-mTOR protein.Conclusion Metformin enhances the drug sensitivity of hepatocellular carcinoma cells to sorafenib,which might be associated with the inhibition of PI3 K/AKT/mTOR signaling and epithelial mesenchymal transformation.
