摘要
目的:探讨灵丹多糖胶囊对CCl_4致小鼠肝纤维化的保护作用。方法:以C57BL/6雄性小鼠为研究对象,构建CCl_4诱导肝纤维化模型,予灵丹多糖胶囊干预。记录实验过程中小鼠体质量治疗变化,检测血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)水平,取肝组织做HE染色,天狼星红染色及α-平滑肌肌动蛋白(α-SMA)免疫荧光染色。结果:与模型组比较,灵丹多糖胶囊各浓度组小鼠血清ALT、AST水平均显著下降(P<0.05,P<0.01),肝脏损伤程度减轻,胶原纤维沉积及α-SMA表达水平均显著下降(P<0.05)。结论:灵丹多糖胶囊能降低肝纤维化小鼠血清ALT、AST水平,改善肝损伤,减少α-SMA的沉淀,对小鼠化学性肝纤维化有保护作用
        Objective: To investigate the protective effect of Lingdan Polysaccharide Capsule against carbon tetrachloride(CCl_4)-induce liver fibrosis injury in mice. Methods: C57 BL/6 male mice were used in this study to duplicate CCl_4-induce liver fibrosis model, intervened with Lingdan Polysaccharide Capsule. The weight of mice and the sterum levels of alanine transaminase(ALT) and aspartate aminotransferase(AST) were examed. The liver paraffin sections were made and tested by the Hematoxylin-eosin(HE) and Sirius Red staining. Furthermore, the deposit of α-smooth muscle actin(α-SMA) in liver tissues was analyzed by immunofluorescent. Results: Compared with the model group, the levels of serum ALT, AST of mice in Lingdan Polysaccharide Capsule group were decreased significantly(P<0.05, P<0.01). The degree of liver injury was alleviated, collagen deposition and the expression of α-SMA were significantly decreased(P<0.05). Conclusion: Lingdan Polysaccharide Capsule could notably lower the mice serum ALT and AST elevation caused by the CCl_4, improve liver tissue injury, and inhibit the generation of α-SMA.
