响应面法优化郁柴茯神颗粒剂的提取工艺

摘要
目的优选郁柴茯神颗粒剂的提取工艺。方法以浸出物得率和HPLC法检测的2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷含量为综合考察指标,单因素筛选提取工艺参数,考察回流时间、乙醇体积分数、乙醇用量和提取次数4个因素,利用Box-Behnken中心组合实验和Design-Expert 8.0.6.1软件统计分析,进一步优化郁柴茯神颗粒剂的提取工艺条件。结果确定郁柴茯神颗粒剂的最佳提取工艺条件为:回流时间为3.0 h,乙醇用量为6倍,乙醇体积分数为80%,提取2次。结论采用响应面法优化郁柴茯神颗粒剂的提取工艺,实际值与预测值吻合度良好,研究结果为郁柴茯神颗粒剂的研究开发提供参考依据。
        Objective To optimize the extraction process of Yuchaifushen Granules.Methods The extraction yield and HPLC method for the determination of 2,3,5,4′-tetrahydroxydistyrene-2-O-β-D-glucoside content were used as the comprehensive evaluation indexes,and the parameters were selected by single factor screening,and 4 factors such as reflux time,ethanol volume fraction,alcohol consumption and extraction times were investigated.Box-Behnken center combined test and software Design-Expert 8.0.6.1 were used to further optimize the extraction conditions of Yuchaifushen Granules.Results The optimum extraction conditions of Yuchaifushen Granules were as follows:the reflux time was 3.0 h,the ethanol dosage was 6 times,and the ethanol volume fraction was 80% with extraction 2 times.Conclusion The response surface method was used to optimize the extraction process of Yuchai fushen Granules.The actual values were in good agreement with the predicted values,and the research results provide reference for the research and development of Yuchaifushen Granules.
