淋病奈瑟菌选择性固体培养基性能评估

摘要
目的对4种商品化淋病奈瑟菌选择性固体培养基(TM培养基)进行性能评估。方法参照《培养基制备指南》及《淋球菌琼脂基础培养基》推荐方法,用生长率、选择性和微生物污染,以及结合临床应用效果,综合评价4种商品化TM培养基的检测性能。结果待测目标菌在进口厂商1(梅里埃)、国产厂商2、3、4TM培养基上的生长率(PR)分别为0.84、0.45、0.30和0.05,其中厂商4 PR低于规定下限,不同厂商之间TM培养基的PR比较,差异均有统计学意义(P<0.05)。4厂商TM培养基对非目标菌的选择因子(SF)为2~5之间,均符合规定。各厂商TM培养基的微生物污染率符合要求(均为0)。调查临床应用厂商1、2、3、4TM培养基的淋病奈瑟菌检出率分别为17.6%、17.0%、7.3%、8.5%,其与相应厂商培养基的PR有较好的一致性。培养基杂菌生长测试,厂商4培养基对真菌防污染能力弱,其他厂商均符合要求。结论市售淋病奈瑟菌固体选择性培养基的质量存在品牌间差异,选用时可通过性能测试选择优质的培养基,以提高临床样本淋病奈瑟菌的检出率。
        Objective To evaluate the performance of four commercial selective media(Thayer-Martin medium,TM)for Neisseria gonorrhoeae.Methods According to the guidelines for the preparation of media and the recommended method of N.Gonorrhoeae agar base medium preparation,the performance of four commercial TM media was comprehensively evaluated by growth rate,selectivity,microbial contamination and clinical application.Results The growth rate(PR)of the tested bacteria on BioMerieux medium and other three domestic manufacturer media were 0.84,0.45,0.30 and 0.05 respectively.Among them,the rate of PRof manufacturer 4 was lower than that of the prescribed lower limit.The PRof TM medium was significantly different among different manufacturers(P<0.05).The selection factor(SF)of all the four TM media for non-target bacteria was from 2 to 5,which met the requirements.The microbial contamination rates of all the tested TM media met the requirements(0).The detection rates of the 1,2,3 and 4 TM media in the clinical settings were 17.6%,17.0%,7.3%and 8.5%respectively,which were in good agreement with the PRof the corresponding manufacturers.The non-target fungi growth test showed that the anti-contamination ability of manufacturer 4 was weak,and other manufacturers met the requirements.Conclusion The quality of the N.gonorrhoeae selective media varies from brand to brand.The high-quality media should be selected by performance test to improve the detection rate of N.gonorrhoeae in clinical settings.
