美国流苏嫩枝扦插及生根内源激素含量的变化

摘要
剪取美国流苏嫩枝作为插穗,研究外源激素、插穗部位、留叶类型等因素对扦插生根的影响,寻求更优的生根处理方式;测定生根进程中内源激素的含量,揭示其与生根的内在关系。结果表明,外源激素3因素对生根率的影响程度为种类>处理时间>浓度,均达到显著差异水平,500mg/L的IBA浸泡60min得到的生根率最高,为56.67%;枝条中部作为插穗时生根率显著高于基部和顶部,为55.00%;插穗保留2片1/2叶时生根率最高,为53.33%,无叶处理的各项指标最差;经IBA处理插穗IAA含量在愈伤组织形成期升高,不定根形成期下降,对照处于不断下降趋势;ABA含量表现为先上升后下降的趋势;GA3、ZR含量在愈伤组织的形成期较高,不定根形成期较低;IBA处理提升了IAA、GA3、ZR含量,降低了ABA含量,有利于插穗生根
        Shoots of Chionanthus virginicus were used as materials to conduct cutting experiment.Effects of exogenous hormones,cutting positions and leaf blade numbers reserved on rooting were examined to seek better rooting treatment.The contents of endogenous hormones during rooting were measured to reveal the intrinsic relationship between the hormones and rooting.The results showed that the influences of three factors related to exogenous hormones on rooting rate was hormone type>treatment time>concentration.The differences among the factors were significantly different.The highest rooting rate(56.67%)was found in the treatment of 500 mg/L IBA soaked for 60 min.The rooting rate of using middle part of shoots as cuttings was 55.00%,significantly higher than those of using shoot base and tip.The highest rooting rate(53.33%)achieved when 2 or 1/2 leaf blades were reserved in the cuttings,and all the indexes without leaf treatment were the worst.The IAA content of the IBA-treated cuttings increased in callus formation period,and decreased in the formation period of adventitious roots,and decreased all the time in the control.The ABA content showed a trend of rising first and then decreasing.The contents of GA3 and ZR were higher in the formation of callus and lower in the formation period of adentitious roots.IBA treatment promoted the contents of IAA,GA3 and ZR in cuttings,reduced the content of ABA,and was conducive to rooting of cuttings.
