苏木乙酸乙酯对急性冠脉综合征外周血CD4+T淋巴细胞增殖及Th17/Treg平衡的影响

摘要
目的观察苏木乙酸乙酯提取液(SAEE)对急性冠脉综合征患者外周血CD4+T淋巴细胞的体外增殖及Th17/Treg比例的影响。方法通过SAEE干预急性冠脉综合征患者外周血单个核细胞,以稳定型心绞痛患者作为对照。运用MTT法检测外周血CD4+T淋巴细胞的增殖,流式细胞术检测Th17、Treg百分比含量,RT-PCR检测白细胞介素-17A(IL-17A) mRNA、叉头翼状螺旋转录因子3(Foxp3) m RNA的表达。结果与稳定型心绞痛患者比较,急性冠脉综合征患者CD4+T淋巴细胞体外增殖能力显著增加,IL-17A的表达显著增加,Foxp3的表达显著降低,Th17/Treg比值显著增高,差异均具有统计学意义(P <0.01);与急性冠脉综合征组比较,SAEE能够显著抑制CD4+T淋巴细胞体外增殖能力,降低IL-17A的表达,升高Foxp3的表达,调控Th17/Treg的比值,其中以苏木乙酸乙酯高剂量组效果最佳,差异均具有统计学意义(P <0.01)。结论SAEE能够通过抑制外周血CD4+T淋巴细胞增殖,调控Th17/Treg的平衡,达到抑制免疫反应和抗动脉硬化的作用。
        Objective: To observe the effect of sppan acetic ether extract on the proliferation of CD4+T lymphocytes and the ratio of Th17/Treg of peripheral blood in patients with acute coronary syndrome. Methods: Sppan acetic ether extract was used to intervene peripheral blood mononuclear cells in patients with acute coronary syndrome,and stable angina pectoris was used as control. MTT assay was used to detect the proliferation of peripheral blood CD4+T lymphocytes,flow cytometry to detect the percentage of Th17 and Treg,and RT-PCT to detect the expression of IL-17 A mRNA and Foxp3 mRNA. Results: Compared with patients with stable angina pectoris,the proliferation ability of CD4+T lymphocytes,the expression of IL-17 A and the ratio of Th17/Treg were significantly increased,and the expression of Foxp3 were significantly reduced(P < 0.01). Sppan acetic ether extract could inhibit the proliferation of CD4 +T lymphocytes in vitro,decrease the expression of IL-17 A,increase the expression of Foxp3,and regulate the ratio of Th17/Treg,compared with acute coronary syndrome group. The effect of Sppan acetic ether extract in high dose group was the best(P < 0.01). All defferences above were statistically significant(P < 0.01). Conclusion: Sppan acetic ether extract can inhibit immune response and anti-atherosclerosis by inhibiting the proliferation of peripheral blood CD4+T lymphocytes and regulating the balance of Th17/Treg.
