江西地区RhD阴性个体的RHD基因与RhCcEe抗原的表达分析

摘要

目的调查江西地区RhD表型确认为阴性者的RhCcEe表型与RHD基因型间的关系,确认RHD基因的外显子模式对RhCcEe抗原表达的影响。方法血清学直接凝集法检测D、C、c、E、e抗原,间接抗人球蛋白试验(IAT)确认RhD阴性血型,对IAT确认为RhD抗原阴性的样本,随机抽取111例,使用PCR-SSP法行RHD基因分型。结果 464例标本中,确认为RhD阴性者451例(97.1983%),RhD变异性13例(2.8017%),111例确认RhD阴性的标本中,共检出六种RHD基因型,包括RHD(-)5例,RHD-CE(2-9)-D 71例,DVI III 1例,DEL RHD1227A 12例,弱D152例,RHD(+)20例,血清学上,111例标本中共检出ccee61例(54.96%),Ccee36例(32.43%),CCee8例(7.21%),CcEe和ccEe各3例(2.70%)。结论 RhD阴性人群中RHD基因序列的不同影响RHCcEe各表型的表达。
        Objective To investigate the relationship between RHD gene and RhCcEe antigen of serologic RhD negtive population in Jiangxi area and find the influnce of the exon model of RHD gene on the expression of RhCcEe antigen. Methods Serological methods were applied to identify Rh antigen-specificity,RhD negative specimens were identified using the IAT(indirect antiglobulin test) Coombs' test. The genetic typing of 111 RhD negative samples was identified based on PCR-SSP. Results A total of 451(97.1983%) RhD negative cases and 13(2.8017%) RhD variants cases were identified among the 464 samples. Six kinds of RHD gene types were detected in 111 cases of RhD negative samples. RHD genetic typing recorded 5 cases of RHD negative,71 cases of RHD-CE(2-9)-D,1 case of D Cat.VI III,12 cases of DEL RHD 1227 A,2 cases of weak D15 and 20 cases of RHD positive. Among 111 RHD gene typing samples,61(54.96%) were ccdee phenotype,36(32.43%) were Ccdee phenotype,8(7.21%)were CCdee phenotype,3(2.70%) were CcdEe and ccdEe seprately. Conclusion The sequence of RHD gene will affect the RhCcEe antigen.
