温胆汤对肥胖痰湿证免疫及炎症细胞因子表达的影响

摘要
目的:探讨祛痰名方温胆汤对高脂饮食诱导的肥胖SD大鼠免疫及炎症细胞因子表达的影响。方法:根据文献方法建立肥胖痰湿证大鼠模型后采用温胆汤灌胃进行干预,比较用药前后大鼠肥胖率、体质量、Lee’s指数、血脂(TG、TC、LDL-C、HDL-C)、免疫T淋巴细胞(CD~+_3、CD~+_4、CD~+_8)及炎症细胞因子(TNF-α、IL-6、IL-17和IL-22)表达的变化。结果:高脂饮食可成功建立肥胖大鼠模型,造模组大鼠肥胖率大于20%,体质量、Lee’s指数明显增高,有显著性差异(P<0.05);温胆汤干预可降低肥胖大鼠肥胖率、体质量和Lee’s指数(P<0.05),改善血脂水平(P<0.05),调整T淋巴细胞CD~+_3、CD~+_4、CD~+_8及CD~+_4/CD~+_8比值的表达,有效调节TNF-α、IL-6、IL-17和IL-22等相关炎症细胞因子的表达(P<0.05)。结论:温胆汤改善肥胖效果明显,其机理可能是通过调控机体免疫机制来改善肥胖炎症反应状态,进而达到纠正肥胖痰湿病理的作用,这为"治痰"名方温胆汤干预肥胖痰湿证提供了科学依据。
        Objective: To investigate the effect of Wendan Decoction on the expression of immune and inflammatory cytokines in obese SD rats induced by high-fat diet.Methods: The obese rat model with syndrome of phlegm-dampness was set up according to the literature method and then gavage administration with Wendan Decoction to compare the obesity rate, body weight, Lee's index, blood lipids(TG, TC, LDL-C, HDL-C), immune T lymphocytes(CD~+_3, CD~+_4, CD~+_8) and inflammatory cytokines(TNF-α, IL-6, IL-17 and IL-22) before and after treatment.Results:High-fat diet may establish successfully obesity rat model, and the obesity rate of the model group was more than 20%.The body weight and Lee's index were significantly increased and there was significant difference compared with those of the normal group(P<0.05).The invervention of Wendan Decoction may reduce obesity rate, body weight and Lee's index(P<0.05), improve blood lipid level(P<0.05), regulate the expressions of CD~+_3, CD~+_4, CD~+_8 and CD~+_4/CD~+_8 ratio of T lymphocytes, and effectively regulate the expressions of TNF-α, IL-6, IL-17 and IL-22 related inflammatory cytokines(P<0.05).Conclusion: Wendan Decoction has obvious effect on improving obesity, and its mechanism may be through regulating the body's immune mechanism to improve the inflammatory state of obesity, and then to correct the pathological effect of obesity with phlegm dampness, which provides a scientific basis for the famous prescription Wendan Decoction for treating phlegm to interfere with obesity phlegm dampness syndrome and then get the role of correcting the phlegm dampness pathology of obesity and provides a scientific basis for Wendan Decoction intervene obesity with syndrome of phlegm-dampness.
