中国人唾液血型物质与FUT2、FUT3基因多态性初步研究

摘要
目的分析36例中国各地区健康随机人群的FUT2、FUT3及唾液ABH血型物质效价,计算FUT2、FUT3基因型检出数量及频率,唾液血型物质效价,比较不同FUT2、FUT3基因之间唾液血型物质的效价。得到FUT2、FUT3基因对于唾液血型物质效价高低的影响,并尝试探索2个基因之间的连锁关系。方法采用中和抑制试验检测唾液中的ABH血型物质。采用基因测序方法检测FUT2、FUT3基因。计算不同FUT2、FUT3基因型唾液血型物质的均值和SD值,使用t检验比较不同FUT2、FUT3基因之间唾液血型物质的效价。结果 36例标本中,FUT2基因测序检出Se Se、Se Sew、SewSew,未发现se基因单倍型。FUT3基因测序检出LeLe、Lele、lele。唾液血型物质效价n分布在-2. 2—15. 5之间(2n)。结论 Se基因对于唾液血型物质效价影响较大,Se基因能够影响个体体液中血型物质的分泌,无论纯合或杂合,影响力无显著差异。而Le基因的影响力远小于Se基因。
        Objective The titers of FUT2,FUT3 and ABH substances in saliva were analyzed in 36 healthy randomized population from different regions in China. Statistical analyzed the Frequency of FUT2,FUT3 genotypes and the ABH substances titration in salivary. Compare the ABH substance titration between the FUT2 and FUT3 genotypes. The effects of FUT2 and FUT3 genes on the ABH substance titration were obtained. Attempts were made to explore the linkage between two genes. Methods Inhibition testing of ABH substance in salivary was performed. The sequencing of FUT2,FUT3 was completed. The mean and SD values of salivary substances in FUT2 and FUT3 were calculated and by t value tests were performed to compare the impact of different FUT2/3 genes on saliva contents. Results In the 36 samples,the FUT2 gene sequencing detected Se Se,Se Sewand SewSewwith no haplotypes of se gene. FUT3 gene sequencing detectedLeLe,Lele and lele. The salivary substance titration was distributed between-2. 2—15. 5( 2 n). Conclusion Se gene has a great influence on the potency of salivary blood group substances. Se gene can affect the secretion of blood group substances in body fluids.No significant difference is found between the effect of homozygous or heterozygous. The Le gene's influence is far less than that of the Se gene.
