摘要
以达乌里胡枝子为对象,研究其没食子酸的最优提取条件,旨在为达乌里胡枝子没食子酸的提取提供依据。结果表明,通过正交试验,达乌里胡枝子没食子酸最佳提取条件为:溶剂浓度60%甲醇,固液比1∶40,超声时间45 min,超声温度60℃;在该条件下,采用高效液相色谱法(HPLC)测定没食子酸的含量(HPLC条件为WaterXBridge C18(250 mm×4.6 mm,5μm),乙腈-0.15%三氟乙酸梯度洗脱,流量为1 m L/min,检测波长为280 nm,柱温30℃),没食子酸在4.91~30.64μg/m L范围内呈现良好的线性关系,平均加样回收率为99.58%,RSD为0.30%;并且利用该方法,测得达乌里胡枝子没食子酸含量为7.57 mg/g。
        The optimal extraction conditions of gallic acid in Lespedeza davurica were studied to provide a basis for the extraction of gallic acid in Lespedeza davurica. The results showed that by orthogonal test, the most suitable condition of extracting gallic acid in Lespedeza davurica was gotten, and that was 60% methanol for solvent concentration, solid-liquid ratio of 1∶40, extraction time of 45 min and temperature of 60 ℃. In this condition, gallic acid content was measured with High Performance Liquid Chromatography(HPLC condition was Water XBridge C18(250 mm×4.6 mm,5 μm), and when gradient elution was carried out with it, the flow of acetonitrile-0.15% trifluoroacetic acid was 1 m L/min by detection wavelength of 280 nm under column temperature of 30 ℃), the gallic acid content within 4.91-30.64 μg/m L showed a good linear relationship, with average recovery of 99.58% and 0.30% RSD. And with this method, the gallic acid content in Lespedeza davurica was measured up to 7.57 mg/g.
