糖心安对Ⅱ型糖尿病大鼠周围神经病变的保护作用及其机理研究

摘要
目的:评价糖心安胶囊对Ⅱ型糖尿病致周围神经病变的防治作用及相关机制。方法:将SD雄性大鼠随机分为两组,即对照组和模型组。所有造模动物灌胃高糖高脂乳,正常对照组给等体积蒸馏水,每天一次,连续4周,然后一次性尾静脉注射链脲菌素25mg/kg以复制2型糖尿病模型。正常对照组动物给予等容积缓冲液。将造模成功大鼠随机分为5组:模型组、盐酸二甲双胍0.1 g/kg+卡托普利0.25g/kg、糖心安0.3 g/kg、糖心安0.15 g/kg、糖心安0.075 g/kg,灌胃给药,模型组和正常对照组分别给于配药溶剂0.5%羧甲基纤维素钠;每周称重,调整给药量,持续给药8周。在末次给药后24小时,测定其坐骨神经运动神经传导速度、血清TG、TC、GLU、SOD、MDA、糖化血清蛋白、AGEs,并取其坐骨神经做病理组织学检测。结果:在造型8周末其模型大鼠血清LDLC含量增加,MDA明显升高,糖化血红蛋白以及糖基化终末产物均显著增加,其坐骨神经传导速度(MNCV)降低,有髓纤维髓鞘空泡变性至严重脱髓鞘,神经纤维变细,部分神经组织局部发生肿胀现象,许旺细胞增殖等病理改变;糖心安0.3g/kg能显著降低血清中MDA、糖化血红蛋白以及AGEs,显著提高坐骨神经传导速度,改善糖尿病大鼠外周神经病理形态学变化。结论:糖心安对Ⅱ型糖尿病周围神经病变具有一定的防治作用,其作用机理可能与其能降低糖尿病大鼠血清糖化血红蛋白、糖基化终末产物以及MDA水平有关,其详细作用机理还有待进一步研究。
        Object: Evaluation of the effect of Tangxinan on diabetic neuropathy( DN). Methods: SD rats were induced diabetic peripheral neuropathy by ig high fat and sugar food and iv STZ 25 mg/kg. 3 days later,DN rats were divided into 5 groups. These rats were ig Tangxinan300 mg/kg,150 mg/kg,75 mg/kg,metformin 100 mg/kg or 0. 5% same volume of CMC respectively for 8 weeks. The levels of TG,TC,GLU,SOD,MDA,HbA 1C and AGEs in serum,the conduction velocity and pathomorphology of sciatic nerve were measured in rats. Results: After8 weeks,the levels of LDL-C,MDA,HbA 1C and AGEs in serum were significantly increased but the conduction velocity of sciatic nerve were significantly decresed in DN rats. Tangxinan0. 3g/kg can significantly reduce the levels of MDA,HbA 1C and AGEs in serum,and improve the conduction velocity and pathomorphology of sciatic nerve in DN rats. Conclusions: These results showed that the protective effect of Tangxinan on the DN may be related to lower the levels of MDA,HbA 1C and AGEs in serum.
