健脾清化方对2型糖尿病大鼠肠道免疫细胞Th1/Th2/Th17/Treg的影响

摘要
目的:研究健脾清化方对2型糖尿病大鼠肠道免疫细胞的影响。方法:采用高脂饮食联合链脲佐菌素(STZ)建立稳定的T2DM大鼠模型,随机分为正常对照组、模型对照组和健脾清化方组,灌胃给药4周,检测大鼠随机及空腹血糖,ELISA法检测空腹胰岛素,流式细胞术检测大鼠肠道免疫细胞Th1、Th2、Th17和Treg比例。结果:与模型对照组比较,健脾清化方降低胰岛素抵抗指数(HOMA-IR),减少小肠组织中免疫细胞Th1、Th2和Th17的细胞比例,增加小肠组织中Treg细胞比例。结论:健脾清化方可改善T2DM大鼠的胰岛素抵抗,其机制可能与调节肠道免疫细胞有关。
        Objective:To explore the effects of Jianpi Qinghua Recipe on intestinal immune cells in type 2 diabetes mellitus(T2DM) rats.Methods:The rat model of T2DM was established by a combination of small dosage of streptozotocin(STZ) injection with high-fat diet.Rats were randomly divided into the control group,model group and Jianpi Qinghua Recipe group.All rats were treated by gavage for 4 weeks.The fasting blood glucose of rats was detected by glucometer instantly.Fasting insulin was detected by ELISA.The proportions of Th1,Th2,Th17 and Treg from rats gut were analyzed by flow cytometry.Results:Compared with the model group,Jiangpi Qinghua Recipe reduced HOMA-IR of T2DM rats,decreased the proportions of Th1,Th2 and Th17 cells and increased the proportion of Treg cells in the small intestine.Conclusion:Jianpi Qinghua Recipe improved insulin resistance in T2DM rats by regulating proportions of Th1,Th2,Th17 and Treg in gut.
