芪灯明目胶囊对自发性糖尿病大鼠视网膜蛋白激酶C及玻璃体管内皮生长因子影响的实验研究

摘要
目的:观察芪灯明目胶囊对自发性糖尿病大鼠视网膜蛋白激酶C及玻璃体管内皮生长因子的影响,探讨中医药治疗糖尿病视网膜病变的机制。
     方法:从形态学等方面对自发性糖尿病大鼠模型进行评价。并采用6个月病程的自发性糖尿病大鼠模型,随机分为6组:阴性对照组、芪灯明目胶囊低、中、高剂量组、递法明组及导升明组,另设有正常对照组。连续灌胃3个月,并于灌胃3月末处死大鼠,通过酶联免疫及免疫组化等方法,观察芪灯明目胶囊对自发性糖尿病大鼠视网膜蛋白激酶C及玻璃体管内皮生长因子的影响。
     结果:(1)自发性糖尿病大鼠(GK大鼠)可以作为人类糖尿病视网膜病变背景期病理改变的模型,特别是Ⅱ型糖尿病动物模型。(2)芪灯明目胶囊有改善自发性糖尿病大鼠生存状态的作用。(3)芪灯明目胶囊具有抑制自发性糖尿病大鼠视网膜蛋白激酶C活化的作用。(4)芪灯明目胶囊可以降低自发性糖尿病大鼠玻璃体管内皮生长因子的水平,从而对新生管的形成起到抑制作用。(5)芪灯明目胶囊对自发性糖尿病大鼠糖有一定降低作用。
     结论:自发性糖尿病大鼠(GK大鼠)可以作为人类糖尿病视网膜病变背景期病理改变的模型,特别是Ⅱ型糖尿病动物模型。芪灯明目胶囊具有抑制自发性糖尿病大鼠视网膜蛋白激酶C活化的作用,降低自发性糖尿病大鼠玻璃体管内皮生长因子的水平,对糖尿病视网膜组织有一定的保护作用,对新生管形成有抑制作用。
Objective: To observe the effect of QiDengMingMu capsule on protein kinase C in retinas and the Vascular endothelial growth factor in vitreous of the experimental animal model of spontaneous diabetic retinopathy rats (Goto-kakizaki rats), and to explore the pathological mechanism of Chinese Medicine on diabetic retinopiathy (DR) .
     Methods: To evaluate the experimental animal model of spontaneous diabetic retinopathy rats (Goto-kakizaki rat) in morphology. The GK rats with six month course are randomly divided into 6 groups according to the weight and the level of blood glucose:model control group, treated groups with high、middle、low dosage of QiDengMingMu capsule , positive control group of Difral and Doxium, and normal control group, respectively. After given drugs or distilled water for 3 months, the rats were put to death and eyeballs of the rats are taken out. Their vitreous and retinas were studied by means of enzyme linked immunosorbent assay (ELISA) and immunohistochemistry respectively.
     Results:(1)The experimental animal model of spontaneous diabetic retinopathy rats may represet background stage of diabetic retinopathy in human, especially the experimental animal model of non-insulin dependent diabetic retinopathy. (2)The Chinese medicine QiDengMingMu capsule has the effect of improving the quality of life of the GK rats. (3)The Chinese medicine QiDengMingMu capsule can inhibit the activity of PKC of the GK rats in retinas. (4)The Chinese medicine QiDengMingMu capsule can reduce the level of the Vascular endothelial growth facter of the GK rats in vitreous. (5)The decrease in the blood glucose level of the GK rats is partly owing to the Chinese medicine QiDengMingMu capsule.
     Conclusion:The experimental animal model of spontaneous diabetic retinopathy rats may represet background stage of diabetic retinopathy in human, especially the experimental animal model of non-insulin dependent diabetic retinopathy. The Chinese medicine QiDengMingMu capsule can the inhibit activity of PKC of the GK rats in retinas and can reduce the level of the Vascular endothelial growth facter of the GK rats in vitreous. So it is a good capillary protective agent to the GK rats'retinal impairment and it can cure the damage of retinas in the GK rats.
