致急性肝胰腺坏死病副溶血弧菌(VP_(AHPND))感染凡纳滨对虾后的动态变化以及定量分析

摘要
致急性肝胰腺坏死病副溶血弧菌(Vibrio parahaemolyticus,VP_(AHPND))是对虾急性肝胰腺坏死病(Acutehepatopancreatic necrosis disease,AHPND)主要病原菌。本研究利用VP_(AHPND)(V-1)对凡纳滨对虾进行浸泡感染试验,探究浸泡感染后VP_(AHPND)在各组织中数量的动态变化。实验设立实验组和对照组,用菌株V-1对实验组浸泡感染,浓度为10^5/mL水体,试验组不进行感染处理,感染后分别于2d-11d采集各组对虾肝胰腺、鳃、肠道、肌肉,实时荧光定量PCR方法检测各组织中VP_(AHPND)数量。实验结果显示,在各取样时间段实验组凡纳滨对虾各组织在均能检测到VP_(AHPND),对照组未检测到VP_(AHPND);实验组对虾肝胰腺组织病理切片显示出VP_(AHPND)的典型症状;最高菌量出现在肝胰腺中,其次是肠道、肌肉、鳃;各组织中VP_(AHPND)数量大致分布在10^1-10^4 copies/mg组织;组织内的菌量随时间大体呈现先上升后下降的趋势。
Vibrio parahaemolyticus(VPAHPND) is the major pathogen is acute hepatopancreatic necrosis disease(AHPND). In this study,Peneaus vannamei were exposed to VPAHPND V-1 by immersion, to explore the dynamic changes of V-1 number in the tissues after immersing infection.The experiment included the experimental group and control group, only to infect the experimental group with the VPAHPND V-1 by immersion at a concentration of 10^5/mL water,no treatment to control group.To collect shrimp hepatopancreas, gill, intestinal and muscle respectively at 2d-11 d after infection, detecting the quantity by qPCR. The experimental results show that the experimental group can detect V-1 in the tissues at all sampling point, not detected in control group; histopathological biopsy showed the shrimp hepatopancreas of experimental group had typical symptoms.
