草鱼几种finTrim基因的生物信息分析

摘要
三重基序(TRIM)蛋白是一类RING锌指结构域、B box结构域、Coiled Coil卷曲螺旋结构域以及其后C端不同结构域组成的大家族。TRIM蛋白在细胞内过程发挥多种功能,包括细胞发育、分化、凋亡和抗病毒免疫反应等。本研究主要针对草鱼中鱼类特有的finTrim基因(ftr51,ftr67,ftr72,ftr82,ftr83,ftr99)进行了分析并预测了其蛋白质结构域,发现这6种基因蛋白质结构域拥有相同的RING锌指结构域(除ftr72外),B box结构域(除ftr67,ftr82,ftr83外)和正向选择发育成的B30.2结构域(除ftr67和ftr72以外),不同结构域说明发挥的功能有所不同。通过系统发育分析显示不同的子集显示家族特异性多样化,因此,说明fintrim基因数量在不同鱼类中有很大的变化。通过对ftr67,ftr82,ftr83,ftr99基因相似率分析发现它们和斑马鱼的相似率很高,同时拥有不同的进化分支,并且进化的速度比人类Trim基因快。
The tripartite motify(TRIM)proteins constitute a large family of protein containing a RING-Bbox-Coiled Coil motify followed by different C-terminal domains. TRIM proteins play multiple roles in various cellular processes, which include cell growth, differentiation, apoptosis and antiviral immunity. We analyzed some novel trim genes in grass carp called finTrim genes,including ftr51, ftr67, ftr72, ftr82, ftr83, ftr99. The protein domain was predicted and formed of nearly identical RING zinc finger domain with the exception of ftr72, B box domain excluding ftr67, ftr82, ftr83, and B30.2 domain that evolved under positive selection excepting ftr67, ftr72.That may demonstrate they involved different cellular processes. A phylogenetic analysis revealed different subsets suggesting lineage-specific diversification events. Therefore, the number of fintrim genes have greatly differences among species in fish. The ftr67, ftr82, ftr83, ftr99 in grass carp closely related to zebrafish, and possess various evolutionary branches, and evolved faster than human.
