棉花TOR信号途径及其与JA信号途径的互作机理研究

摘要
棉花尤其陆地棉(Gossypium hirsutum L.)是全世界范围内纺织用纤维的最主要来源,是最重要的经济作物之一,在我国国民经济的发展中发挥至关重要的作用。环境胁迫是影响棉花产量的重要因素,影响棉花生长发育、降低棉花产量。雷帕霉素作用靶标(Targetofrapamycin,TOR)信号途径是调控真核生物生长和发育的关键信号途径,在真核生物中普遍存在。茉莉酸(Jasmonic acid,JA)信号途径在调节植物应对胁迫中发挥极其重要的作用。解析TOR与JA信号途径之间的互作机理,为增强植物抵御外界胁迫的能力,减少作物产量损失奠定理论基础。本研究以陆地棉为对象,发现TOR抑制剂以及MeJA能够协同抑制棉花幼苗的生长,且TOR抑制剂处理后棉花内源JA含量显著上升。此外,TOR抑制剂处理下,JA合成及信号传导的关键基因的转录发生显著改变,本研究初步研究了TOR及JA信号途径之间的互作。
Cotton,especially upland cotton(Gossypium hirsutum L.),provides the major resource of textile fiber worldwide,is one of the most important cash crops and plays a key role in the development of national economy.Environmental stress significantly influences cotton yield.To deal with environmental stresses,crops like cotton frequently encounter yield penalties resulting from effects of environmental stresses on development.Target of rapamycin(TOR) signaling pathway functions as a key regulator of growth and development,and widely exists in eukaryotes.Jasmonic acid(JA),as an important plant hormone,extensively involves in response to abiotic and biotic stress.Dissecting the underlying genetic components and the mechanistic in sights of TOR-JA crosstalk in plants may provide a strategy to improve tolerance and resistance to environmental stress and reduce production loss,which has a great significance to crop production.In the present study,we found that TOR inhibitor and MeJA inhibit cotton growth synergistically,and TOR inibitor treatments significantly increase.endogenous JA content.Transcriptome analysis also revealed that the transcript levels of genes involved in JA biosynthesis and signal transduction were changed under TOR inhibition.This study makes a preliminary study of the crosstalk between TOR and JA signaling pathways in cotton.
