月季杂交幼胚培养技术研究

摘要
为了有效缩短蔷薇属植物的杂交育种周期,本试验以窄叶藤本月月红(R.chinensis‘Narrow-leaflet Climber')为母本,月月粉(R.chinensis‘Old Blush')为父本进行人工杂交,研究了早期杂交胚培养的技术程序,分析了胚龄对胚培养效果的影响,比较了幼胚培养和传统的杂交种子育苗方式获得杂交后代苗的效率。结果表明,该杂交组合适宜幼胚培养的最早胚龄为授粉后90天左右,采用幼胚培养方式,从杂交授粉到获得杂种后代的时间较常规方式缩短了109天(31.48%),SSR分子标记分析证明胚培养获得的后代为真杂种。在此基础上,建立了月季花品种间杂交幼胚培养的技术程序。
To shorten the period of cross breeding in genus Rosa,in our experiment,immature embryos of R.chinensis' Narrow-leaflet Climber' × R.chinensis' Old Blush' were in vitro cultivated,and the effects of embryo age on seed germination was analyzed.The results showed that the seeds of more than 90 days old germinated successfully with gemination rate 16.67—23.53%.The seedlings were identified to be true hybrids with SSR mrkers analysis.Compared to traditional hybridization procedure,the breeding period decreased by 31.4%(109 d) with 90-day-old embryos culture method.Comprehensive analysis of different factors,immature embryo culture was considered an ideal way to improve hybridization breeding efficency.
