口服葡萄糖对团头鲂幼鱼血液呼吸爆发、肝脏抗氧化酶活性及PI3K/Akt/Nrf2通路关键调控因子相对表达量的影响

摘要
为探究口服高糖(3g/kg,鱼重)对团头鲂幼鱼血液呼吸爆发,肝脏抗氧化酶活性及PI3K/Akt/Nrf2通路关键因子相对表达量的影响,选用初重为(19.94±0.03g)的鱼随机分组进行试验。结果发现,口服后血糖出现波动,在1h和12h时显著升高(p<0.05,下同),且1h时伴有血细胞数目显著降低,肝脏中T-SOD等关键抗氧化酶活性显著升高,抗氧化应激相关通路基因PI3K、Akt、SIRT1、Nrf2、HO-1相对表达量极显著上调(p<0.01);12h时,血糖的再次升高,导致血液呼吸爆发活性显著下降,伴有血清蛋白含量的显著降低。本研究结果表明,口服高糖导致团头鲂幼鱼出现血糖异常波动,呼吸爆发功能受抑制;血糖升高诱导肝脏PI3K/Akt/Nrf2信号通路关键因子及下游抗氧化应激蛋白相对表达量上调,以抵抗氧化应激。
A trial was conducted to investigate the effects of oral glucose(3g/kg, bw) on the blood respiratory burst, hepatic enzyme activities and gene expression of PI3K/Akt/Nrf2 pathway in juvenile blunt snout bream M.amblycephala(19.94±0.03g). The results indicated that blood glucose significantlyincreased(p<0.05) at 1hand12 hafter oral, blood cell counts significantly decreased, hepatic antioxidant enzyme activities significantly increased, and relative expression of PI3 K, Akt, Nrf2, HO-1 very significantly(p<0.01) up regulated at 1h. With increase ofblood glucose at 12 h, blood respiratory burst and protein content significantly decreased. In conclusion,oral glucose trial result in abnormal fluctuation of plasma glucose, inhibition of respiratory burst,and increase of plasma glucose up regulated antioxidative stress related pathway.
