Differential Expression of miRNA-125b and Validation of Predicted Target Genes in the Skin Tissue of Fine-wool Sheep and Cashmere Goats

MicroRNAs(miRNAs) is a new class of endogenous small non-coding RNA which can negatively regulate gene expression via pairing to 3'-untranslated region(UTR) of targeted mRNAs to degrading targeted mRNA or repressing mRNA translation.This study predicted and validated the target gene of miRNA-125 b in the skin tissue of fine-wool sheep and cashmere goats.The target genes of miR-125 b were predicted by miRGen and DAV1 D software.The MAX Dimerization Protein 4(MXD4) and Fibroblast growth factor receptor 2(FGFR2) were chosen as target genes as they correlated with hair follicle development.The expressions of miR-125 b,MXD4 and FGFR2 were examined by qPCR(Real-time Quantitative PCR) and western blot in skin tissue of fine-wool sheep and cashmere goats and Human Embryonic Kidney(HEK) 293 T cells which overexpressed miR-125 b.The luciferase reporter gene system was used to verify the correlation between miR-125 b and MXD4,FGFR2 gene.The expressions of MXD4 and FGFR2 in the skin of cashmere goats was significantly higher than that in fine-wool sheep(p<0.01) and miR-125 b expression level was on the contrary(p<0.01).The levels of MXD4 and FGFR2 could be down-regulated by miR-125 b in HEK 293 T cells.Furthermore,the target sequences of miR-125 b were existed in 3'UTR of FGFR2 and MXD4 gene.MXD4 and FGFR2 are target genes of miR-125 b and miR-125 b may play a regulatory role in the development of sheep hair follicle through these two target genes.
