不同LED光质处理对莲瓣兰组培苗生长的影响

摘要
本研究以播种的莲瓣兰‘大雪素'萌发后生长14个月的无菌苗为培养材料,探究了不同光质的发光二极管(LED)组合对‘大雪素'组培快繁过程中根状茎增殖,新芽分化及其生根率的影响。结果表明:相对于普通荧光作为光源,450~480nm蓝光照射可显著提高莲瓣兰增殖培养过程中根状茎增殖效率;630nm红光作为光源可显著提高继代培养过程中莲瓣兰新生芽生长速率;而在壮苗生根培养过程中,使用红光和蓝光混合搭配作为光源可促进根长生长,并以红光和蓝光1:3比例处理效果最佳,生根数目较对照组增加近2倍,叶片数目增加30%以上。可见,合理搭配使用LED光源作为光源,对莲瓣兰‘大雪素'的组织培养具有重要意义。
This study examined the effect of the red and blue light-emitting diode(LED) light on the proliferation of the rhizomes,the differentiation of cespitose buds and the rooting rate of the tissue culture seedlings of Cymbidium Lianpan.14-month-old seedlings of Cymbidium Lianpan ' Da xue su ' were used as the explants.The results showed that illuminating of the blue light of 450 — 480 nm significantly improved the rhizome proliferation efficiency during the proliferation culture,compared with common fluorescence.The red light of 630 nm markedly increased the growth rate of the cespitose buds in the process of subculture.However,the mixture of the red and blue light used as the light source facilitated the expansion of the root length,especially when treated with the red and blue light by the proportion of 1:3,with almost 2 times-increasing of the striking roots number compared to the control group.All of our results indicated that the LED illumination system used in proper proportion of red and blue light is very important for the tissue culture of Cymbidium Lianpan.
