优化微柱离心法提高脂质体包封率测定的准确性

摘要
目的:优化脂质体包封率测定的条件,提高脂质体包封率测定结果的准确性。方法:以积雪草苷为模型药,建立HPLC法测定积雪草苷含量;采用薄膜分散法分别制备空白脂质体与载药脂质体,利用微柱离心法分离脂质体与游离药物。分别将空白脂质体与纯化后的载药脂质体以及两者的稀释混悬液过微柱,比较脂质体的过柱率,最终采用权重系数对脂质体的包封率进行修正。结果:在所选定的分离条件下,空白脂质体的过柱率为95%,纯化的载药脂质体的过柱率只有90%,二者存在明显差异(P<0.05);脂质体稀释后,空白脂质体的过柱率无明显变化,而载药脂质体的过柱率则从89.49%降至74.25%。结论:两种测定方法脂质体的过柱率差异较大,采用权重系数修正包封率,测得结果更为准确。
Objective To optimize the conditions for determination of the encapsulation efficiency of liposomes and improve the determination accuracy.Methods:To establish HPLC method for determine the contents of asiaticoside,which was used as the active pharmaceutical ingredient.The liposomes were prepared by the thin film dispersion method and separated from free drug by micro-column centrifugation method.Then,the study compared the recovery rate through micro-column between blank liposomes,and purified liposome swas proceeded,and both of them diluted suspensions were so on,respectively.At last,the weight coefficients was used to correct the encapsulation efficiency.Results:Under the separation conditions,the recovery rate of blank liposomes was 95%while the recovery rate of purified liposomes was only 90%,there was a significant difference(P <0.05);when the two kinds of liposomes were diluted,the recovery rate of blank liposomes has no changes,but the recovery rate of purified liposomes has decreased from 89.49%to 74.25%.Conclusion:It is showed that the recovery rate of liposomes has great differences in the two methods.The encapsulation efficiency would be more accuracy with weight coefficient corrected.
